Description

The polyclonal antibody against ipaD can be generated by immunizing a rabbit with the recombinant invasin ipaD protein (1-332aa) from Shigella flexneri. Once collected from the rabbit serum, the ipaD antibody is further purified using protein G. This specific ipaD antibody is ideal for detecting the ipaD protein in Shigella flexneri through ELISA and WB applications.

The ipaD protein, a component of the type III secretion system (T3SS) of Shigella flexneri, plays a crucial role in the pathogenesis of Shigella flexneri by promoting invasion of the host intestinal epithelial cells. IpaD acts as a gatekeeper protein, controlling the secretion and translocation of other effector proteins through the T3SS. It also helps in the formation of a pore complex that allows the bacteria to enter the host cell.

Target Background

Function
Required for bacterial invasion of host cells. Controls IpaB and IpaC secretion, and the efficiency with which they are physically inserted into target cell membranes. These proteins are exported via TTSS to form a pore in the host membrane that allows the translocation of the other effectors into the host cytoplasm. Along with IpaB, is essential for both blocking secretion through the Mxi/Spa translocon in the absence of a secretion-inducing signal, and for controlling the level of secretion in the presence of this signal.
Gene References into Functions
  1. IpaD, the putative needle-tip protein of the S. flexneri type III secretion system, has been crystallized; in-drop proteolysis resulted in the production of several new crystal forms. PMID: 16946465
  2. IpaD forms part of the structure at the needle tip in Shigella flexner and antibodiesblock entry of bacteria into epithelial cells. PMID: 17110044
Subcellular Location
Secreted. Note=Secreted via the type III secretion system (TTSS). Localizes to the tip of the external secretion needle that is part of the TTSS apparatus.
Protein Families
