The Human Transforming Growth factor β3 (TGFB3) ELISA kit is specifically designed to standardize the measurement of TGFB3 in biological samples, including serum, plasma, and cell culture supernates, for research and biomedical laboratories. This kit has been validated with various criteria, including sensitivity, specificity, precision (less than 10%), linearity, recovery, and inter-batch difference. The assay principle applied in this kit is the quantitative Sandwich enzyme immunoassay. The microtiter plate has been pre-coated with an anti-human TGFB3 antibody. Standards or samples are added to the appropriate microtiter plate wells with the Biotin-conjugated TGFB3 antibody. Next, HRP-Avidin is added to each microplate well. After TMB substrate solution is added, wells containing TGFB3, Biotin-conjugated TGFB3 antibody, and HRP-conjugated Avidin will exhibit a color change. The enzyme-substrate reaction is terminated by the addition of the stop solution and the color intensity is measured via a microplate reader at 450nm. The levels of TGFB3 in the samples is determined by comparing the O.D. (optical density) of the samples to the standard curve.

TGFB3 is a pleiotropic growth factor exclusively expressed in mammals and mainly secreted and stored as a latent complex in the extracellular matrix (ECM). TGFB3 plays a role in the maturation of myoepithelium, inhibition of milk secretion, and the epithelial-mesenchymal transition (EMT) process. It is essential for the development of the palate and lungs and functions mainly by regulating epithelia-mesenchymal interactions during embryonic, fetal, and neonatal development. Additionally, TGFB3 may also participate in the process of wound closure, orchestrating an orderly migration of dermal and epidermal cells in damaged skin.

  • A micro ELISA plate --- The 96-well plate has been pre-coated with an anti-human TGFB3 antibody. This dismountable microplate can be divided into 12 x 8 strip plates.
  • Two vials lyophilized standard ---Dilute a bottle of the standard at dilution series, read the OD values, and then draw a standard curve.
  • One vial Biotin-labeled TGFB3 antibody (100 x concentrate) (120 μl/bottle) ---Act as the detection antibody.
  • One vial HRP-avidin (100 x concentrate) (120 μl/bottle) ---Bind to the detection antibody and react with the TMB substrate to make the solution chromogenic.
  • One vial Biotin-antibody Diluent (15 ml/bottle) ---Dilute the Biotin-antibody.
  • One vial HRP-avidin Diluent (15 ml/bottle) ---Dilute the HRP-avidin solution.
