Intra-assay Precision (Precision within an assay): CV%<8%
Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision (Precision between assays): CV%<10%
Three samples of known concentration were tested in twenty assays to assess.
To assess the linearity of the assay, samples were spiked with high concentrations of rat CYP7A1 in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

Target Background

(From Uniprot)
Catalyzes a rate-limiting step in cholesterol catabolism and bile acid biosynthesis by introducing a hydrophilic moiety at position 7 of cholesterol. Important for cholesterol homeostasis.
Gene References into Functions
  1. lnc-HC negatively regulates cholesterol metabolism within hepatocytes through physical interaction with hnRNPA2B1. By further binding to the target messenger RNA of Cyp7a1 or Abca1, the lnc-HC-hnRNPA2B1 complex decreases expressions of the two genes that are implicated in cellular cholesterol excretion. PMID: 26663205
  2. Administration of glucocorticoids (GCs) enhanced liver bile acid uptake by increasing the expression of the major hepatocyte basolateral bile transporter (Ntcp) in rats. The reduced expression of a bile acid synthesis rate-controlling enzyme, Cyp7a1, suggests that treatment with GCs suppressed hepatic bile acid synthesis. PMID: 27133208
  3. Dietary fats and glucomannan plus spirulina added to pork significantly alters Cyp7a1 expression in Zucker rats. PMID: 26475369
  4. Ginsenosides promoted liver cholesterol metabolism due to upregulation of CYP7A1. PMID: 23108811
  5. Portal blood FGF15/19 levels remain unchanged while the corresponding ileal mRNA and liver CYP7A1 levels change significantly in cholesterol fed rats. PMID: 23852734
  6. Capsaicinoids consumption down-regulated the mRNA levels of HMG-CoA reductase and CYP7A1 in the cholesterol-free diet group but up-regulated the CYP7A1 level in the cholesterol-enriched diet group. PMID: 23617777
  7. insulin-dependent suppression results in impairment of the hepatic cholesterol/bile acid metabolisms PMID: 21817852
  8. Maternal protein restriction elevates cholesterol in adult rat offspring due to repressive changes in histone modifications at the cholesterol 7alpha-hydroxylase promoter PMID: 21372147
  9. Cytochrome P450 7A1 cholesterol 7alpha-hydroxylation: individual reaction steps in the catalytic cycle and rate-limiting ferric iron reduction. PMID: 21147774
  10. essential for heart and skeletal muscle function PMID: 20215401
  11. The mRNA and protein expressions of SHP and CYP7A1 were increased in livers of pregnant rats with intrahepatic cholestasis. PMID: 18578998
  12. Cyp7a1 activity is modified after chronic ingestion of depleted uranium in the rat. PMID: 20362056
  13. The increased recruitment of LXRalpha, a Cyp7a1 stimulatory pathway, and decreased expression of FGF15 and phosphorylated Erk1/2, a Cyp7a1 repressive pathway, combined to increase Cyp7a1 expression during lactation. PMID: 19957370
  14. LXR alpha is the dominant regulator of CYP7A1 transcription PMID: 12054605
  15. transcription suppressed by alpha1-antitrypsin peptide via interaction with AFP PMID: 12223476
  16. Data reveal a fundamental difference in the regulation of CYP7A1 in rodent and human hepatocytes. PMID: 12554795
  17. ARP-1/COUP-TFII acts as a transcription activator of CYP7A1. PMID: 12777384
  18. results suggest that the mechanisms of regulation of these bile acid metabolizing enzymes are significantly different during liver regeneration PMID: 16081591
  19. These results suggest that the cyp7A1 gene plays an important role in development of hypercholesterolemia in Prague hereditary hypercholesterolemic rats. PMID: 18226361
  20. Increased Cyp7a1 expression inlactating rats provides a mechanism for the increase in the bile acid pool. PMID: 18292185
  21. The effect of osthole on protein expression of PPARalpha/gamma and related target molecules such as CYP7A and DGAT protein expression in the liver of hyperlipidemic fatty liver rats. PMID: 18696335
  22. A relationship between the decreased expression of hepatic CYP7A1 mRNA and protein and dilated ER may contribute to hypercholesterolemia in chronic hypothyroidism. PMID: 19788618
