Description

R-spondin-1 is a critical potentiator of Wnt/β-catenin signaling, and this recombinant human RSPO1 demonstrates strong functional activity with an ED50 of 1–10 ng/ml in a TOPFlash reporter assay using HEK293T cells, confirming its capacity to robustly activate canonical Wnt pathway readouts. Covering the full-length mature protein (aa 21–263) with a C-terminal 6xHis tag, this construct is expressed in mammalian cells to preserve native glycosylation patterns essential for proper folding and receptor engagement with LGR4/5/6. This validated bioactivity supports use in intestinal and other adult stem cell organoid expansion, osteogenic differentiation studies, and receptor-ligand binding assays such as SPR or ELISA-based competition experiments. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based proliferation and survival assays as well as in vivo regenerative medicine models requiring stringent endotoxin control.

Activator of the canonical Wnt signaling pathway by acting as a ligand for LGR4-6 receptors. Upon binding to LGR4-6 (LGR4, LGR5 or LGR6), LGR4-6 associate with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. Also regulates the canonical Wnt/beta-catenin-dependent pathway and non-canonical Wnt signaling by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway. Acts as a ligand for frizzled FZD8 and LRP6. May negatively regulate the TGF-beta pathway. Has a essential roles in ovary determination. Regulates Wnt signaling by antagonizing DKK1/KREM1-mediated internalization of LRP6 through an interaction with KREM1.
