Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Please provide the following information regarding this protein (all expression systems).
1)As, this protein is His-tagged, can we use this antigen as ELISA standard ?
2)Will antibody bind with same affinity to the tagged and untagged antigen ?(as if due to tagging, affinity will vary, then his-tagged protein can’t be used as standard for the untagged protein)(please provide any supportive data for the same if available)
3)If we need same protein without any tag, will you be able to provide it ?

① Pls be noted that the Tag of this EP protein is N-terminal 6xHis-SUMO-tagged.
The recombinant proteins are usually used as ELISA standard, in theory, this protein can also be used as the ELISA standard.
However, it is also necessary to see whether the protein matches the antibody in the ELISA. We haven't verified about this for the time being, so we can't 100% guarantee, we recommend you to purchase a small size to have a try first.
② In combination with the your description, we think you want to ask whether the tagged antigen and the untagged antigen have an effect on the affinity of the antibody.
This needs to be determined by combining with the immunogen information of the corresponding antibody and purification method.
a. If the immunogen of your antibody is a fusion protein, purified using proteinA/G or purified by fusion protein affinity chromatography, then choosing untagged antigen as the standard will reduce antibody affinity.
b. If the immunogen of your antibody is a fusion protein, which is purified by untagged target protein affinity chromatography, the antibody affinity is the same for the untagged or tagged/fusion protein.
c. If the immunogen of your antibody is an untagged protein, using proteinA/G purification or antigen affinity chromatography, choosing tagged/fusion protein as the standard will reduce antibody affinity.
Generally, it's better to detect endogenous samples (natural samples). If you want the data to be as accurate as possible, so it is best to use untagged protein for immunization and using untagged protein as the standard.
③ If you need the untagged protein, we can provide tag removal service:
Please remark your requirement for tag removal if you need the untagged protein or tag removal when placing the order. We can try enzyme digestion, but we can't guarantee 100% successfully.The overall success rate of enzyme digestion data analysis is 75%-86%.
* Not all protein tags can be removed as some proteins will be very unstable after tag removal.
* If we succeed in removing the tag, we will charge for extra cost.
* If we fail in removing the tag, we will provide the fusion protein and won’t charge for any extra cost of tag removal, and remark this information in datasheet as follows
“Note: The laboratory determined that the Tag on your protein could not be removed with standard laboratory procedures. Your protein is being supplied with the Tag intact.”
* Generally, the delivery time will be extended for 3 days.
PS: If the tag removal result of the current engineered vector is not good, we will change another vector.

Target Background

Function
Minor protein of the capsid that localizes along the inner surface of the virion, within the central cavities beneath the L1 pentamers. Plays a role in capsid stabilization through interaction with the major capsid protein L1. Once the virion enters the host cell, L2 escorts the genomic DNA into the nucleus by promoting escape from the endosomal compartments and traffic through the host Golgi network. Mechanistically, the C-terminus of L2 possesses a cell-penetrating peptide that protudes from the host endosome, interacts with host cytoplasmic retromer cargo and thereby mediates the capsid delivery to the host trans-Golgi network. Plays a role through its interaction with host dynein in the intracellular microtubule-dependent transport of viral capsid toward the nucleus. Mediates the viral genome import into the nucleus through binding to host importins. Once within the nucleus, L2 localizes viral genomes to host PML bodies in order to activate early gene expression for establishment of infection. Later on, promotes late gene expression by interacting with the viral E2 protein and by inhibiting its transcriptional activation functions. During virion assembly, encapsidates the genome by direct interaction with the viral DNA.
