Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
May be required to maintain the proliferative capacity of stem cells. Stabilizes MDM2 by preventing its ubiquitination, and hence proteasomal degradation.
Gene References into Functions
  1. Nucleostemin expression participates in the preservation of the viability and integrity of embryonic stem cells. PMID: 25522312
  2. nucleostemin protects hematopoietic stem cells (HSCs) from DNA damage accumulation and is required for the maintenance of HSCs. PMID: 24140061
  3. Mechanistic studies performed in mouse embryonic fibroblast cells showed that loss of nucleostemin triggers DNA damage and growth arrest independently of the p53 status or rRNA synthesis PMID: 23798389
  4. NS plays a critical role in regeneration of both hepatic precursor cells and hepatocytes in response to liver injury. PMID: 22775537
  5. Nucleostemin in the maintenance of ESC self-renewal, demonstrate the importance of rapid transit through G1 for this process, and expand the known classes of reprogramming factors. PMID: 22689653
  6. NS functions downstream of Myc as a rate-limiting regulator of cell proliferation and transformation. PMID: 22081066
  7. NS undergoes a ubiquitin- and MDM2-independent proteasomal degradation when intracellular GTP levels are markedly reduced PMID: 22318725
  8. Reactive oxygen species regulate nucleostemin oligomerization and protein degradation. PMID: 21242306
  9. Findings establish the tumor-initiating and molecular features of NS-enriched mammary tumor cells, suggesting that NS may offer a valuable therapeutic target. PMID: 21045149
  10. skeletal muscle cell differentiation has a unique expression profile of NS in that it is continuously expressed during differentiation. PMID: 19914205
  11. Data show that the NS-GFP system provides a powerful tool to elucidate stem cell biology in normal and malignant tissues. PMID: 19805150
  12. analysis of nucleolar localization of stem cell protein nucleostemin PMID: 15857956
  13. Nucleostemin has a unique ability, derived from an ancestral function, to control the proliferation rate of stem/progenitor cells in vivo independently of p53. PMID: 17000755
  14. Has roles in establishing early embryogenesis and delaying cellular senescence of embryonic fibroblasts. PMID: 17000763
  15. Transgenic mice were created that expressed green fluorescent proteins from the genomic locus of nucleostemin using the bicistronic or the ATG-fusion approach. PMID: 17941047
  16. Nucleostemin expression in cardiomyocytes is induced by fibroblast growth factor-2 and accumulates in response to Pim-1 kinase activity. PMID: 18519946
  17. NS is a marker of undifferentiated status in the germ cell lineage during prepubertal spermatogenesis PMID: 18802033
  18. Loss of nucleostemin results in reduced cell proliferation and increased apoptosis in both embryonic stem (ES) cells and ES cell-derived neural stem/progenitor cells. PMID: 19418458
