Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Metalloprotease with deubiquitinase activity that plays important regulator roles in hematopoietic stem cell function, blood cell production and immune response. Participates in the normal programming of B-cell responses to antigen after the maturation process. Within the cytoplasm, plays critical roles in the repression of innate immunity and autoimmunity. Removes 'Lys-63'-linked polyubiquitins from TRAF3 and TRAF6 complexes. Attenuates NOD2-mediated inflammation and tissue injury by promoting 'Lys-63'-linked deubiquitination of RIPK2 component. Suppresses the CGAS-STING1 signaling pathway by cleaving STING1 'Lys-63'-linked ubiquitin chains. In the nucleus, acts as a hematopoietic transcription regulator derepressing a range of genes essential for normal stem cell differentiation including EBF1 and PAX5 in B-cells, ID2 in NK-cell progenitor or FLT3 in dendritic cell precursors. Deubiquitinates monoubiquitinated histone H2A, a specific tag for epigenetic transcriptional repression, leading to dissociation of histone H1 from the nucleosome.
Gene References into Functions
  1. Mysm1 is a critical epigenetic regulator of epidermal development and ESC specification through interference with p53-mediated apoptosis and cell cycle inhibition-and potentially modification of p63-regulated programs-whereas ESC maintenance later in adult life may be compensated through yet to be investigated mechanisms. PMID: 29495602
  2. Mysm1 enhanced function of the IRF2 and IRF8 promoters, suggesting that Mysm1 governs the IRFs for hematopoietic stem cell homeostasis. PMID: 27277682
  3. MYSM1 activity at the prepro-B cell stage of development is important for the normal programming of B cell responses to stimulation once they complete their maturation process. PMID: 27895164
  4. Overall we conclude that Mysm1 expression within the niche is not essential for the maintenance of hematopoietic homeostasis. PMID: 27833034
  5. this study shows that MYSM1 is involved in the maintenance, activation and survival of CD8+ T cells PMID: 28066899
  6. The results demonstrate that MYSM1 plays a critical role in Mesenchymal Stromal Cell maintenance and differentiation. This study also underscores the biological significance of deubiquitinase activity in Mesenchymal Stromal Cell function. PMID: 26915790
  7. MYSM1 is a critical negative regulator of p53 transcriptional programs in hematopoiesis, and its repression of Bbc3/PUMA expression is essential for multipotent progenitor survival, and partly contributes to maintaining hematopoietic stem cell function PMID: 26768662
  8. this study uncovers a critical role for MYSM1 in epigenetically repressing plasma cell differentiation and antibody production, in addition to its opposing, active role in B cell development. PMID: 26348977
  9. MYSM1 is essential for normal HSC function and progression of hematopoiesis in the fetal liver and HSC maintenance in adult bone marrow. PMID: 26125289
  10. 2 motifs in MYSM1 are essential for innate immune suppression: the SWIRM domain that interacts with TRAF3 and TRAF6 and the metalloproteinase domain that removed K63 polyubiquitins. MYSM1 is a key negative regulator of the innate immune system PMID: 26474655
  11. Loss of hematopoietic stem cell function and bone marrow failure in Mysm1-knockout mice are the result of p53 activation. PMID: 25710881
  12. Data indicate that histone H2A deubiquitinase Mysm1 is required for dendritic cell development. PMID: 25217698
  13. Mysm1-mediated epigenetic regulation has a role in controlling hematopoietic stem cell maintenance, self-renewal, and differentiation PMID: 24014243
  14. MYSM1 is required for natural killer cell maturation, including the the recruitment of nuclear factor Il-3 to the ID2 locus. PMID: 24062447
  15. Data indicate a role for histone H2A-deubiquitinase Mysm1 in the maintenance of bone marrow stem cell function, in the control of oxidative stress and genetic stability in hematopoietic progenitors. PMID: 22184403
  16. MYSM1 derepresses EBF1 transcription in B cell progenitors by orchestrating histone modifications and transcription factor recruitment to the EBF1 locus. PMID: 22169041
