Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Plays a critical role in the sodium-dependent reabsorption of bile acids from the lumen of the small intestine. Plays a key role in cholesterol metabolism.
Gene References into Functions
  1. data support the concept that under physiological conditions microbiota stimulate Gata4, which suppresses Asbt expression PMID: 26022694
  2. In Asbt-deficient mice, the lack of bile acid uptake in the distal small intestine, which we recently documented using novel in vivo imaging results in more than 10-fold increased fecal bile acids PMID: 26210740
  3. ileal ASBT protein is degraded by a ubiquitin-dependent pathway in response to enterobacteria-associated bile acids PMID: 23872411
  4. Ileal Fibroblast Growth Factor-15 (FGF15) expression was significantly reduced in Asbt(-/-)apoE(-/-) mice and was inversely correlated with expression of hepatic cholesterol 7-hydroxylase (Cyp7a1). PMID: 23880190
  5. The functional importance of proline and uncharged polar residues in the highly conserved region of mouse Slc10a2 was determined. PMID: 23374508
  6. Enterobacteria-mediated bile acid biotransformation modulates intestinal bile acid transport via downregulation of SLC10A2 expression. PMID: 20884752
  7. These data confirm that bile acids and upregulation of ASBT play a crucial role in NEC pathogenesis and suggest that inhibition of ASBT could be utilized as a therapeutic modality against this disease. PMID: 20616306
  8. Slc10a2 is essential for efficient intestinal absorption of bile acids PMID: 12819193
  9. ASBT-mediated bile salt uptake in the distal ileum might also modulate intestinal fluid secretion PMID: 16037545
  10. The lithogenic diet induced biliary hyperplasia and reduced bile salt transporter Slc10a2 expression in C57L mice. PMID: 18717763
  11. Demonstrate a posttranscriptional modulation of ileal ASBT function and membrane expression by phorbol ester via a PKCzeta-dependent pathway. PMID: 19571234
  12. Highly conserved region contains residues involved in the substrate interaction, function, and cellular localization of mSlc10a2. PMID: 19584562
