Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Involved in the targeting and/or fusion of transport vesicles to their target membrane during transport of proteins from the early endosome to the lysosome. Required for heterotypic fusion of late endosomes with lysosomes and homotypic lysosomal fusion. Required for calcium regulated lysosomal exocytosis. Involved in the export of chylomicrons from the endoplasmic reticulum to the cis Golgi. Required for exocytosis of mediators during eosinophil and neutrophil degranulation, and target cell killing by natural killer cells. Required for focal exocytosis of late endocytic vesicles during phagosome formation.
Gene References into Functions
  1. Spastin regulates VAMP7-containing vesicles trafficking in cortical neurons. PMID: 28392418
  2. VAMP7 mediates fusion of BLOC-1-dependent transport carriers with melanosomes, illuminate SNARE recycling from melanosomes as a critical BLOC-3-dependent step. PMID: 27482051
  3. VAMP7-mediated release of interleukin-12 at the immune synapse is a mechanism to transmit innate signals to T cells. PMID: 26972013
  4. VAMP7 mediates plasma membrane fusion of vesicles containing TRPM8. VAMP7-deficient mice exhibit reduced functional expression of TRPM8 in sensory neurons and concomitant deficits in cold avoidance and icilin-induced cold hypersensitivity. PMID: 26843440
  5. VAMP-7(-/-) platelets demonstrated a partial defect in dense granule exocytosis and impaired aggregation. Consistent with a role in cytoskeletal remodeling, spreading on matrices was decreased in VAMP-7(-/-) platelets compared to wild-type controls. PMID: 25999457
  6. VAMP7 controls the phosphorylation of Lat, formation of the TCR-Lat signaling complex and, ultimately, activation of T cells. PMID: 23666293
  7. The binding of VAMP7 to delta-adaptin requires the VAMP7 SNARE motif to be engaged in SNARE complex formation and hence AP3 must transport VAMP7 when VAMP7 is part of a cis-SNARE complex. PMID: 22521722
  8. Behavioral characterization studies indicate that deletion of Vamp7 exon 7 is associated with increased anxiety in mice. PMID: 22323709
  9. VAMP7 is required in neurons to extend axons to the full extent. VAMP7 does not seem to be required for epithelial cell polarity and lysosomal exocytosis. PMID: 21740490
  10. VAMP7, and other synaptic vesicle proteins thus target to both recycling and resting pools. However, the proportions differ dramatically, with higher levels of VGLUT1 and VAMP2 in the recycling pool and of VAMP7 in the resting pool PMID: 21835344
  11. VAMP7 controls exocytosis of proteolipid protein (myelin) 1 from late endosomal/lysosomal organelles as part of a transcytosis pathway PMID: 21490207
  12. Data suggest that tetanus neurotoxin-insensitive vesicle-associated membrane protein TI-VAMP defines a new pathway of membrane delivery required for optimal Fc receptor-mediated phagocytosis [TI-VAMP]. PMID: 15470500
  13. The results revealed a marked defect in neurite outgrowth and arborization, suggesting that Ca(2+)-dependent, Syt VII-regulated exocytosis of late endosomes/lysosomes plays a role in the addition of new membrane to developing neurite extensions. PMID: 16641243
  14. Study demonstrates that the clathrin-mediated endocytosis of the SNARE VAMP7 is directly mediated by Hrb, a clathrin adaptor and ArfGAP. PMID: 18775314
