Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Target Background

Function
Receptor for netrin required for axon guidance. Mediates axon repulsion of neuronal growth cones in the developing nervous system upon ligand binding. NTN1/Netrin-1 binding might cause dissociation of UNC5C from polymerized TUBB3 in microtubules and thereby lead to increased microtubule dynamics and axon repulsion. Axon repulsion in growth cones may also be caused by its association with DCC that may trigger signaling for repulsion. Might also collaborate with DSCAM in NTN1-mediated axon repulsion independently of DCC. Also involved in corticospinal tract axon guidance independently of DCC. Involved in dorsal root ganglion axon projection towards the spinal cord. It also acts as a dependence receptor required for apoptosis induction when not associated with netrin ligand.

  1. T835M (rs137875858) in UNC5C increased hippocampal neuronal cell death and susceptibility to neurotoxic stimuli. PMID: 25419706
  2. Both cvd and hob mutations are predicted to lead to truncated UNC5H3 proteins lacking their cytoplasmic region required for Netrin-Unc5h3 signaling pathway. Therefore, we conclude that Unc5h3 is causative gene of both cvd and hob mutant phenotypes. PMID: 15010202
Involvement in disease
Defects in Unc5c are the cause of cerebellar vermis defect (cvd) and hobble (hob) phenotypes. Cvd and hob rats exhibit cerebellar and midbrain defects, possibly as a result of abnormal neuronal migration, and exhibit laminar structure abnormalities in the fused cerebellar hemispheres and ectopic cerebellar tissues in the cerebello-pontine junction.
