Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Inhibitor of PPP1CA. Has over 1000-fold higher inhibitory activity when phosphorylated, creating a molecular switch for regulating the phosphorylation status of PPP1CA substrates and smooth muscle contraction.
Gene References into Functions
  1. an impairment of the rapid CPI-17 signaling pathway mediating alpha1A-adrenoceptor activation is a cause of hypo-responsiveness of splanchnic resistance arteries, as an end organ of orthostatic blood pressure reflex, in the orthostatic hypotension model rats PMID: 28717991
  2. Emodin attenuates tonic tension through the blockade of PKCdelta and CPI-17-mediated MLC-phosphatase inhibition. PMID: 24909118
  3. Rac1 coordinates with RhoA to increase myosin light chain (MLC) phosphorylation by inactivation of CPI-17/MYPT1 phosphatase, which decreases MLC phosphatase activity thus promoting MLC phosphorylation and cell contraction PMID: 25502873
  4. Nuclear accumulation of CPI-17 is detected in proliferating vascular smooth muscle cell cultures and in neointima cells of a vascular injury model. PMID: 23541585
  5. results suggest that myocardin-independent Sp1/Sp3 pathways regulate the CPI-17 transcription through multiple kinases, such as ERK1/2, p38, JNK, PKC, and ROCK in response to environmental changes PMID: 22538237
  6. a novel signaling cascade that links RhoA-mediated calcium sensitivity to MEF2-dependent myocardin expression in VSMCs through a mechanism involving p38 MAPK, PP1alpha, and CPI-17. PMID: 22275376
  7. Data show that receptor stimulation increases Ca(2+) sensitivity of contractile elements through CPI-17 phosphorylation via the PKC/ROCK pathways and MYPT1 phosphorylation via the ROCK pathway in intestinal smooth muscle. PMID: 22004286
  8. The mechanism of the effect of estradiol on the expression of G-family proteins in vitro and in vivo is reported. PMID: 20046026
  9. distribution of mouse and rat PPP1R14A was more specific and different from that of humans PMID: 11931393
  10. ILK may activate smooth-muscle contraction both directly, via phosphorylation of myosin, and indirectly, via phosphorylation and activation of CPI-17 and PHI-1, leading to inhibition of myosin light-chain phosphatase PMID: 12144526
  11. phosphorylated by p21-activated kinase PMID: 12359219
  12. Agonists induce PHI-1 phosphorylation in VSMCs and divergent kinase signaling couples agonist stimulation to PHI-1 and CPI-17 phosphorylation. PMID: 16267107
  13. RhoA/ROCK/CPI-17 pathway is considered to play an important role in the ET-1-induced Ca(2+) sensitisation of bronchial smooth muscle contraction. PMID: 17071121
  14. CPI-17 expression is reversibly controlled in response to the phenotype transition of smooth muscle cells that restricts the signal to differentiated smooth muscle cells and particular cell types. PMID: 19437030
