Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Component of the pheromone signal transduction pathway. It mediates pheromone signals acting between STE20 and STE11. It is absolutely required for pheromone-induced transcription of FUS1. May play a role in cell-cycle arrest in response to pheromone.
Gene References into Functions
  1. defining regions of Ste5 that mediate nuclear export PMID: 26824509
  2. Structural determinants of the specificity of a membrane binding domain of the scaffold protein Ste5 of budding yeast: implications in signaling by the scaffold protein in MAPK pathway. PMID: 22285780
  3. findings show that the scaffold protein Ste5, which organizes the yeast mating mitogen-activated protein kinase pathway, does not use sequestration to prevent misactivation of the mating response; Ste5 appears to use a conformation mechanism PMID: 22878499
  4. The signaling proteins Ste5 and Ste20 have recognition sequences, or "docking" sites, for G1/S (interphase) cyclins. These docking sites bind preferentially to the cyclin Cln2, and strongly enhance Cln2-driven in vivo substrate phosphorylation. PMID: 21945277
  5. a switch-like dissociation of Fus3 from Ste5 that is necessary to generate the switch-like mating response PMID: 20400943
  6. KSS1 is activated by Ste5. PMID: 15192700
  7. Ste5p scaffold feedback phosphorylation is controlled by an associated MAPK cascade PMID: 15322134
  8. Ste5 scaffold has a more active role in the yeast mating pathway; a fragment of Ste5 allosterically activated autophosphorylation of Fus3 PMID: 16424299
  9. Ste5 contains a pleckstrin-homology (PH) domain (residues 388-518) that is essential for its membrane recruitment and function. PMID: 16847350
  10. Cln/CDK disrupts Ste5 membrane localization by phosphorylating a cluster of sites that flank a small, basic, membrane-binding motif in Ste5; effective inhibition requires multiple phosphorylation sites and a substantial accumulation of negative charge PMID: 17289571
  11. Fus3 activity requires the scaffold protein Ste5; when binding to Ste5 is abrogated, Fus3 behaves like Kss1, and the cells no longer respond to a gradient or mate efficiently with distant partners PMID: 18538663
  12. study found that the subcellular localization of Ste5 and cell cycle progression have dramatic effects in controlling the stability of Ste5 PMID: 19001089
  13. The dual requirement for both Ste7 and this Ste5 domain in Fus3 activation explains why Fus3 is selectively activated by the mating pathway and not by other pathways that also utilize Ste7. PMID: 19303851
