Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Binds to the C-terminal PTS1-type tripeptide peroxisomal targeting signal (SKL-type) and plays an essential role in peroxisomal protein import.
Gene References into Functions
  1. Cysteine-specific ubiquitination protects the peroxisomal import receptor Pex5p against proteasomal degradation. PMID: 26182377
  2. Surface plasmon resonance analysis revealed a bivalent interaction mode for the phosphorylated PTS1 receptor Pex5 and Pcs60. PMID: 26359497
  3. The membrane-associated import receptor Pex5p together with its docking partner Pex14p forms a gated ion-conducting channel which can be opened to a diameter of about 9 nm by the cytosolic receptor-cargo complex. PMID: 20154681
  4. Data suggest that the N-terminal half of Pex5p facilitates the import of the major matrix protein Fox1p, and can be functionally replaced by Pex18p. PMID: 15456864
  5. identified ubiquitinated forms of Pex5p in deletion mutants of those PEX genes that have been implicated in recycling of Pex5p from the peroxisomal membrane into the cytosol PMID: 15536088
  6. Pex5p monoubiquitination in wild type cells is required for the recycling of Pex5p from the peroxisome, but Ubc4p-mediated polyubiquitination of Pex5p in mutants may function as a disposal mechanism for Pex5p when it gets stuck in the import pathway PMID: 15632140
  7. Results demonstrate that the AAA peroxins mediate the ATP-dependent dislocation of the peroxisomal targeting signal-1 (PTS1) receptor from the peroxisomal membrane to the cytosol. PMID: 16007078
  8. a translocation-competent state of the PTS1 receptor (Pex5p) enters the membrane via protein-lipid interactions before it tightly associates with other peroxins PMID: 16849337
  9. Study demonstrated that either poly- or monoubiquitination of Pex5 is required for the ATP-dependent release of the protein from the peroxisomal membrane to the cytosol as part of the receptor cycle. PMID: 17452527
  10. Pex4p-dependent ubiquitination of Pex5p occurs on a cysteine residue PMID: 17550898
  11. E2D1/2/3 (UbcH5a/b/c) are the mammalian functional counterparts of yeast/plant Pex4p PMID: 18359941
  12. The RING domain of Pex10p exhibits E(3) ligase activity in vitro, in combination with the human E(2) enzyme UbcH5a, a homologue of ScUbc4p, but not when ScPex4p is used as an E(2) enzyme in the reaction. PMID: 18644345
  13. Pex2 mediates the Ubc4-dependent polyubiquitination whereas Pex12 facilitates the Pex4-dependent monoubiquitination of Pex5. PMID: 19687296

Subcellular Location
Cytoplasm. Peroxisome membrane; Peripheral membrane protein. Note=Its distribution appears to be dynamic. It is probably a cycling receptor found mainly in the cytoplasm and as well associated to the peroxisomal membrane through a docking factor (PEX13).
