Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Protein kinase that acts both on serines and on tyrosines. It acts as a dosage-dependent negative regulator of entry into mitosis (G2 to M transition). Phosphorylates and inhibits cdc2.
Gene References into Functions
  1. Regulation of Wee1 localization around the spindle pole body during the G2/M transition is important for proper mitotic entry and progression. PMID: 21233285
  2. in fission yeast Wee1 is the major inhibitory kinase regulating CDK activity in S phase. PMID: 26918805
  3. This cell cycle arrest is dependent on the Wee1 tyrosine kinase inhibiting the key cell cycle regulator, CDK1/Cdc2. Mutant cells lacking Wee1 are not arrested at G2 upon glucose limitation and lose viability faster than the wild-type cells under glucose-depleted quiescent conditions, suggesting that this cell cycle arrest is required for extension of chronological lifespan PMID: 26804466
  4. Wee1 phosphorylates Cdc2 Tyr15 which leads to replication slowdown in the fission yeast under nitrosative stress. PMID: 26006103
  5. Dnt1 regulates G2/M transition and cytokinesis by downregulating Wee1 kinase. PMID: 24006256
  6. Substitution of a conserved methionine residue to phenylalanine in Plo1, Orb5 and Wee1 kinases increased the sensitivities to ATP analogues. PMID: 23986474
  7. Data show that the protein Skb1 interacted with both Cdr1 and the Cdr1 inhibitory target Wee1. PMID: 23615447
  8. Sid2 activation midway through G2 phase promotes Fin1 phosphorylation and mitotic commitment throught pom1/Wee1 cell geometry network. PMID: 22684255
  9. in fission yeast Cpc2/RACK1 positively regulates from the ribosome the mitotic onset by modulating both the protein levels and the activity of Wee1 PMID: 20974849
  10. The changed division response (Cdr) kinases Cdr1 and Cdr2 are negative regulators of Wee1 and they are required for the Tra1-dependent alterations to Wee1 function. PMID: 20194963
  11. findings demonstrate that G2 arrest induced by HIV-1 Vpr is also associated with increase in phosphorylation & amount of Wee1 in a Chk1/Cds1-independent manner; Rad24 appears to contribute to Vpr-induced G2 arrest by elevating Wee1 expression PMID: 16968670
