ACE2 Antibodies

ACE2 (angiotensin-converting enzyme 2), a homolog of ACE, is a monocarboxypeptidase that counteracts the activation of the renin-angiotensin-aldosterone system (RAAS), thus regulating the systemic blood pressure and renal function and alleviating adverse cardiac remodeling and cardiac dysfunction. Besides, ACE2 also functions as a receptor for SARS-CoV and SARS-CoV-2 and facilitates the invasion into the host cell of both SARS viruses. Anti-ACE2 antibodies may be beneficial to study the pathology of and cardiovascular diseases and SARS-CoV-2 infection.

CUSABIO offers 15 rabbit anti-ACE2 polyclonal antibodies produced from rabbits immunized with diverse immunogens from multiple species. They can react with human, mouse, rat, Saccharomyces, and Schizosaccharomyces. The ACE2 antibodies in the following catalog have features as follows:

High specificity and good sensitivity. CUSABIO's ACE2 antibodies have high specificity and good sensitivity for recognization and detection of ACE2. And these ACE2 antibodies seldom cross-reacted or interfered with their analogs.

Various immunogens. These antibodies are generated from rabbits immunized with multiple immunogens from human, rat, Saccharomyces cerevisiae, and Schizosaccharomyces pombe, including recombinant fragment corresponding to amino acids 395-638 of human ACE2, synthesized peptide derived from the internal region of human ACE2, a fusion protein of human ACE2, recombinant Saccharomyces cerevisiae ACE2 protein, recombinant Schizosaccharomyces pombe ACE2 protein, and a peptide sequence corresponding to amino acids 105-118 of rat ACE2.

Wide species reactivity. The ACE2 antibodies manufactured by CUSABIO can react with multiple species, including humans, mice, rats, Saccharomyces, and Schizosaccharomyces.

Validated for multiple applications. The ACE2 antibodies have been validated for ELISA, WB, IHC, and IF.

High purity. The purity of partial ACE2 antibodies is greater than 95% (Antigen Affinity-purified & Affinity purified or Protein G purified). There are four purification methods for the ACE2 antibodies: Protein G purification, Protein A purification, Antigen Affinity purification, and Affinity purification. Sometimes two of the four purification methods are combined to purify a product.

Labeled with multiple antibody markers. Some of the ACE2 antibodies are conjugated with labeling molecules such as HRP, FITC, and Biotin. These labeled ACE2 antibodies can be used for localization, qualitative & quantitative analysis, purification of corresponding ACE2 antigens. ACE2 Background

Angiotensin-converting enzyme 2 (ACE2) is a type I integral membrane glycoprotein [1]. ACE2 is expressed in most tissues, and its highest expression is found in the kidney, the endothelium, the lungs, and heart [1] [2]. It contains a single catalytic metallopeptidase unit in the extracellular domain, which is partially homolog with that of ACE [2]. Its C-terminal domain is not catalytic and shares a 48% identical sequence with collectrin [3], which may interpret ACE2's function as a carboxypeptidase. ACE2 and its homolog collectrin have been identified as essential molecules for the expression of neutral amino acid transporters on the surface of epithelial cells. Although ACE2 is a homolog to ACE, conventional ACE inhibitors can not inhibit ACE2 activity. Calmodulin- binding sites within the cytoplasmic tail of ACE2 may influence the uncoating of the catalytic ectodomain. As a strong negative regulator in the renin-angiotensin system (RAS), ACE2 counteracts the activity of ACE. By targeting angiotensin II, ACE2 has shown protective effects in the cardiovascular system and many other organs. Additionally, ACE2 was found to act as an important receptor of human coronaviruses such as SARS-CoV and SARS-CoV-2. The human coronaviruses enter the host cell through the binding of viral spike glycoprotein (S) to the ACE2 on the host cell membrane and the S priming by serine protease TMPRSS2 [4]. So ACE2 becomes a crucial therapeutic target for COVID-2019.
