Ephrin (Eph family receptor interacting proteins) belongs to the Ephrin family and is a ligand for the Eph receptor. The ephrin ligands are divided into two subfamilies, ephrinA (ephrin A1-A6) and ephrinB (ephrin B1-B3). The difference between the two subfamilies is that the ephrinA subgroup is an extramembrane protein anchored to the surface of the cell membrane by a glycosylphosphatidylinositol chain (GPI), whereas the ephrinB subgroup is a transmembrane molecule with an extracellular Eph binding region and a short cytoplasmic region.
EFNA5 is a gene encoding a protein called Ephrin-A5, which belongs to the ephrinA subgroup. Ephrin-A5 can bind to EphA2, A3, A4, A5, A7, A8, B2.
With binding to different molecules, EFNA5 shows various function. For example, EFNA5 activates the EPHA3 receptor to regulate cell–cell adhesion and cytoskeletal organization; binding to the receptor EPHA2, EFNA5 may regulate lens fibre shape and interactions, and may be key for maintaining lens transparency; interacting with EPHA5, EFNA5 mediates communication between pancreatic islet cells to regulate glucose-stimulated insulin secretion. Additionally, Ephrin-A5 played a role in pathologic conditions such as tumors.
Ephrin mediates intercellular signal transduction by binding to Eph. The Eph/Ephrin signaling pathway is outlined as follows: Eph receptors undergo self-phosphorylation upon activation, and then activate signaling molecules in downstream signaling pathways such as Ras, Rae, Rho, Rap1, and FAK to regulate various biological effects of cells through corresponding signaling pathways.
Eph-ephrin signaling controls cell localization during normal and carcinogenic development by regulating cell adhesion and de-adhesion, thereby affecting cell development. It is also critical to the embryological process, especially the development of the nervous system. Eph-ephrin signaling regulates neural crest cell migration, axonal orientation, and neuroangiogenesis during embryogenesis and development.
