TNFRSF13C is a gene providing an instruction of making a protein named tumor necrosis factor receptor superfamily member 13C (usually abbreviated as TNFRSF13C). The protein encoded by this gene is also called B-cell-activating factor receptor, BAFF receptor (BAFF-R), BLyS receptor 3 or CD268. BAFF-R is a BAFF-specific type II transmembrane protein, which only binds specifically to BAFF and cannot bind to any other ligands of the TNF family, including APRIL.
Similar to the classic signal transduction pathway of the TNF family, BAFF-R also activates the downstream signal pathway by interacting with ligands and then adapting to the downstream linker molecule TNFR-associated factor (TRAF), which eventually leads to proliferation of cells. BAFF-R specifically interacts with TRAF3, negatively regulates and activates the downstream non-canonical NF-κB pathway, and further mediates the survival of B cells.
The BAFF-BAFFR signaling pathway plays an important role in the proliferation, differentiation and maturation of B cells. The imbalance of the activity of the BAFF-BAFFR signaling pathway can cause the immune imbalance of the body, resulting in a series of lesions such as tumorigenesis, autoimmune diseases and post-transplant rejection.
