Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Plasma membrane anion exchange protein of wide distribution.
Gene References into Functions
  1. results strongly suggest that Ae2 is a critical factor in the innate response to C. albicans. This study represents an important contribution to a better understanding of how Dectin-1 expression and fungal clearance is regulated. PMID: 27391897
  2. Data indicate a key functional role of anion exchanger 2 (AE2) in the maintenance of the normal physiology of biliary epithelial cells (BEC) and the pathogenic consequences of reduced AE2 expression, including abnormal intrinsic characteristics of BEC and their production of signal molecules that lead to the chronic inflammatory responses in small bile ducts. PMID: 27592379
  3. AE2 deficiency results in intrahepatic T-cell activation and PD-1/PD-L1 mediated deletion, favoring autoimmune cholangitis. PMID: 26396175
  4. These findings demonstrate that CD8(+) T cells are critically dependent on Ae2 for pHi homeostasis and tuning of cell proliferation and activation. Ae2 thus constitutes a novel target to modulate CD8(+) T-cell responses. PMID: 24515893
  5. Rescue of SLC4A2-deficient osteoclasts with functionally defined mutants of SLC4A2 indicates regulation of actin cytoskeletal reorganization by anion-exchange activity and intracellular pH, independent of SLC4A2's long N-terminal cytoplasmic domain. PMID: 23341620
  6. expression of Slc4a2 significantly decreased in enamel organs from Mmp20(-/-) mice. A feedback mechanism regulates ion-responsive gene expression during enamel development. PMID: 20929715
  7. AE2 is a component of the basolateral ion transport mechanisms that support anion secretion in the proximal colon. PMID: 20110461
  8. GFP-tagged Ae2 localization depends upon MAPK activity PMID: 19823673
  9. results are consistent with the hypothesis that AE2 mediates the major portion of the exchange across the basolateral membrane of mouse parotid and sublingual acinar cells PMID: 12958022
  10. Male Ae2-/- mice are infertile, with an interruption of spermiogenesis. PMID: 14673081
  11. AE2 is essential for gastric acid secretion and for normal development of secretory canalicular and tubulovesicular membranes in mouse parietal cells PMID: 15123620
  12. importance to pH-dependent regulation of AE2 activity of the surrounding region of aa 312-578 PMID: 15452108
  13. there are regulatory determinants in the AE2 N-terminal cytoplasmic domain of AE2c1 PMID: 16286476
  14. Knockout mice, which suffer from severe hemolytic anemia and spherocytosis, display cardiac hypertrophy and impaired cardiac function. PMID: 17056673
  15. contribution of nonconserved charged residues within the C-terminal transmembrane domain of AE2 to regulation by pH through mutation to the corresponding AE1 residues PMID: 17492309
  16. Basolateral bicarbonate secretion via the anionic exchanger Ae2 is essential for mineral growth in the maturation stage enamel. PMID: 18042363
  17. Mouse strain-specific coding polymorphism in the Slc4a2/Ae2 gene encodes Ae2c2 variants differing in isoform-specific dominant negative activity. PMID: 18192337
  18. These results highlight the importance of bicarbonate transport via Ae2 in maintaining pH(i) homeostasis in cultured mouse fibroblasts. PMID: 18319251
  19. AE2 is expressed at the apical membrane. Transcripts for AE2a were detected in RNA isolated from cultured ameloblast-like LS8 cells. AE2 is expressed in ameloblasts in vivo in a polarized fashion. PMID: 18362326
  20. Ae2 deficiency alters pH(i) homeostasis in immunocytes and gene expression profile in cholangiocytes, leading to immunologic and hepatobiliary changes that resemble PBC. PMID: 18471521
  21. data highlight SLC4A2 as a critical mediator of osteoclast differentiation and function in vitro and in vivo. PMID: 18971331
  22. critical segments of RL1 constitute part of an AE2 pH sensor that, together with residues within the N-terminal half of the TMD, constrain the AE2 polypeptide in a conformation required for regulation of anion exchange by pHi. PMID: 19103596
  23. Severe osteopetrosis was encountered in Ae2 knockout (Ae2-/-) mice where the skeletal development was impaired with a higher diffuse radio-density on x-ray examination and the bone marrow cavity was occupied by irregular bone speculae PMID: 19164575
  24. In contrast to the abnormalities observed in the long bones, the skull of Ae2(a,b)(-/-) mice showed no alterations, indicating that calvaria osteoclasts may display normal resorptive activity. PMID: 19564250
