Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
RNA-binding protein, which is essential for gametogenesis in both males and females. Plays a central role during spermatogenesis. Acts by binding to the 3'-UTR of mRNA, specifically recognizing GUU triplets, and thereby regulating the translation of key transcripts.
Gene References into Functions
  1. The 3'-UTR-dependent suppression of DAZL in mouse follicular oocytes is required for the progression of normal pre-implantation development. PMID: 29883445
  2. Down-regulated expression of Sox9 and Dazl may play important roles in MBP-induced testis injury. PMID: 28496037
  3. findings demonstrate that DAZL associates with mRNA of Tet1, a catalyst of 5-hydroxylation of methyl-cytosine, and enhances Tet1 mRNA translation; DAZL enhances TET1-mediated cytosine hydroxymethylation in embryonic stem cells that are actively reprogramming to a pluripotent ground state PMID: 26077710
  4. Both yeast and mouse contain an RNA-binding protein, Rim4 and DAZL, respectively, that functions in translation and forms SDS-resistant aggregates during gametogenesis. PMID: 26411291
  5. Our findings strengthen the importance of RA and Dazl in the meiotic transition, provide important details about the Stra8 pathway, and open avenues to investigate early meiosis through analysis of Rec8 induction and function PMID: 25102060
  6. Data indicate that the germ cell-specific RNA binding protein DAZL limits both pluripotency and somatic differentiation in nascent primordial germ cells (PGCs). PMID: 25418731
  7. demonstrate that Boll is also transiently expressed during oogenesis in the fetal mouse ovary, but is simultaneously co-expressed within the same germ cells as Dazl PMID: 24086306
  8. Studies indicate that in Dazl-Knockout mice, cord/tubule-like structures, OSE invaginations and clusters of steroidogenic cells became increasingly prominent with age, but these abnormal structures remained benign. PMID: 23417416
  9. alternative splicing of exon8 of mDazl leads to the generation of the novel splice variant, Dazl-Delta8. PMID: 23298641
  10. DAZL was translocated to stress granules upon heat stress. Furthermore, stress granule assembly was significantly diminished in the early male germ cells of Dazl-knockout mice. DAZL induced the sequestration of signaling molecules such as RACK. PMID: 22223682
  11. structures suggest that the number of DAZL molecules bound to GUU triplets in the 3'-UTR provides a method for modulating the translation of a target RNA PMID: 22021443
  12. If DAZL expression is reduced, then follicles can grow to the late follicular stage despite declining levels of circulating FSH, leading to more follicles ovulating and increased litter size. PMID: 21270429
  13. Licensing of gametogenesis, dependent on RNA binding protein DAZL, as a gateway to sexual differentiation of fetal germ cells. PMID: 21504946
  14. CPEB1 regulates Dazl post-transcriptionally, and that DAZL is essential for meiotic maturation and embryonic cleavage PMID: 21460039
  15. Stra8 and Dazl are highly expressed in premeiotic germ cells that do not undergo further differentiation in aged mouse ovaries. PMID: 20157580
  16. Dazl contributes to the differentiation of embryonic stem cells into germ cells. PMID: 20008336
  17. Dazl is a master gene controlling germ cell differentiation and that ectopic expression of Dazl promotes the dynamic differentiation of mouse ES cells into gametes in vitro. PMID: 19783541
  18. Mouse DAZL protein can bind to 3'UTR of mTex19.1 mRNAs and may repress mTex19.1 expression at the translational level.Confirmed by both electrophoretic mobility shift assay and translation assay in Zebrafish embryo detecting the luciferase activity. PMID: 19247806
  19. Data show that EBs expressed Oct-4, OTR, OT, and DAZL. PMID: 19695304
  20. regulates a subset of mRNAs necessary for germ cell development and cell cycle progression PMID: 11804965
  21. mouse Dazl has the ability to stimulate translation and function at the level of translation initiation. PMID: 16001084
  22. Dazl stimulates translation via the Mvh 3'-UTR. Dazl-mediated regulation of Mvh translation is crucial for mammalian spermatogenesis PMID: 16278232
  23. Dazl is required as early as E12.5-E13.5, shortly after its expression is first detected, and that inbred Dazl-/- mice of C57BL/6 background provide a reproducible standard for exploring Dazl's roles in embryonic germ cell development. PMID: 16310179
  24. Sycp3 is a biologically relevant target of Dazl-mediated translation in mammals. PMID: 17526644
  25. Protein expression of a Dazl gene during embryogenesis in mice. PMID: 17761180
  26. Dazl gene may play different roles in human and mouse spermatogenesis by regulating different members of the downstream gene family. PMID: 18452650
  27. study shows that extrinsic & intrinsic factors regulate meiotic initiation in embryos; DAZL, which is expressed by postmigratory germ cells, is a key intrinsic factor, enabling those cells to initiate meiosis in response to retinoic acid PMID: 19074348
  28. report establishes the translational regulator Dazl as a component of pluripotency, genetic, and epigenetic programs at multiple time points of germ cell development. PMID: 19468308
