Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Ligand-gated chloride channel which is a component of the heteropentameric receptor for GABA, the major inhibitory neurotransmitter in the brain. Plays an important role in the formation of functional inhibitory GABAergic synapses in addition to mediating synaptic inhibition as a GABA-gated ion channel. The gamma2 subunit is necessary but not sufficient for a rapid formation of active synaptic contacts and the synaptogenic effect of this subunit is influenced by the type of alpha and beta subunits present in the receptor pentamer. The alpha2/beta2/gamma2 receptor exhibits synaptogenic activity whereas the alpha2/beta3/gamma2 receptor shows very little or no synaptogenic activity.
Gene References into Functions
  1. Gabra2-1 mice display spontaneous seizures during development, accompanied by early mortality PMID: 30087324
  2. At a lower concentration of fluoxetine and its active metabolite norfluoxetine (that is in the therapeutic range), study observed anxiogenic- and depressive-like actions in alpha2 knock-out mice, but not in alpha2+/+ mice, suggesting that in 129x1/SvJ mice a decrease in alpha2-containing GABAARs promotes an increased sensitivity to the prodepressant-like effects of chronic fluoxetine treatment. PMID: 28587819
  3. Findings suggest that ovarian cycle-related progesterone and neurosteroids regulate alpha2-GABA-A receptor expression in the hippocampus via a non-progesterone receptor pathway, which may be relevant to menstrual-cycle related brain conditions. PMID: 28137424
  4. Results show that the expression levels of Gabra2, as well as of GABA receptor-mediated inhibitory neurotransmission, are reduced in Shank2 e6-7, but not in e7 KO mice compared with their own wild type littermates. Shank2 e6-7 KO mice treated with allosteric modulator for the GABAA receptor reverses spatial memory deficits, indicating that reduced inhibitory neurotransmission may cause memory deficits in Shank2. PMID: 27544825
  5. role for: (i) H101 in species-typical binge-like drinking, (ii) Q241 in escalated chronic drinking, and (iii) S270 and/or L277 in the development of forced abstinence-associated social deficits PMID: 27717041
  6. Pharmological manipulation by clobazam, a common anticonvulsant with preferential affinity for the GABRA2 receptor, revealed dose-dependent protection against hyperthermia-induced seizures in Scn1a+/- mice. These findings support Gabra2 as a genetic modifier of the Scn1a+/- mouse model of Dravet syndrome. PMID: 27768696
  7. Results indicate that 174 proteins were associated with gamma-aminobutyric acid receptor subunit alpha 2 (pHalpha2). PMID: 27044742
  8. whilst not directly involved in reward perception, the alpha2- subunit may be involved in modulating the "energising" aspect of cocaine's effects on reward-seeking. PMID: 24481569
  9. Wild-type and mutant mice have similar baseline nociceptive sensitivities and develop similar hyperalgesia specifically targeting alpha2-GABAA receptors which exert their antihyperalgesic effect through enhanced spinal nociceptive control. PMID: 24045508
  10. GABRA2 subunit expression in hippocampus modulates neurogenesis through nuclear factor of activated T cell (NFATc)4 activity. PMID: 24948817
  11. Disrupting the clustering of GABAA receptor alpha2 subunits in the frontal cortex leads to reduced gamma-power and cognitive deficits. PMID: 24043839
  12. Recovery of function following alcoholic intoxication is regulated by Gabra2 and Gabra3 receptor subunits. PMID: 23147414
  13. These findings suggest that alpha2-containing GABA(A) receptors expressed in the CA3 region provide the inhibition that controls hippocampal rhythm during cholinergically induced oscillations PMID: 23109109
  14. The findings of this study implicated alpha2 and alpha3 subunit containing GABA(A) receptors as key mediators of the reward-related effects of benzodiazepines PMID: 22763624
  15. The results of this study emphasize the critical role of GABAergic synaptic signaling for structural maturation of adult-born GCs and formation of glutamatergic synapses. PMID: 22745509
  16. Data suggest that the availability of distinct GABA(A) R subtypes (Gabra2, -4, and Gabrd) provides a molecular mechanism endowing spatiotemporal specificity to GABAergic control of neuronal maturation in adult brain. PMID: 21722213
  17. Data show that L. rhamnosus (JB-1) reduced GABA(Aalpha2) mRNA expression in the prefrontal cortex and amygdala, but increased GABA(Aalpha2) in the hippocampus. PMID: 21876150
  18. presynaptic GABAA receptors residing on the axons or terminals of primary nociceptors play a significant role in spinal pain control. PMID: 21632935
  19. Arc and Fos, increased about 250% in the alpha2 mutants, Behavioral effects (loss of righting reflex, rotarod) of flurazepam and pentobarbital were not different between alpha2 mutants and wild-type, but they were enhanced for alpha1 knock-in mice. PMID: 21056629
  20. results show that alpha 2-containing GABAA receptors represent a specific molecular substrate that can regulate depressive-like states. PMID: 20965216
  21. alpha2-containing GABA(A) receptors are important for specific behavioral effects of ethanol that may be relevant to the genetic linkage of this subunit to alcoholism. PMID: 20876231
  22. activation of alpha2-GABA(A) receptors is both necessary and sufficient for behavioral sensitization. PMID: 20133874
  23. Hybridization signals for the alpha2 mRNAs first appeared in the differentiating zone at embryonic day 13 (E13). And was detected in the migrating and differentiating granule cells and cerebellar nucleus neurons until postnatal day 14 (P14). PMID: 14766194
