Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Plays a role in the regulation of glomerular permeability, acting probably as a linker between the plasma membrane and the cytoskeleton.
Gene References into Functions
  1. While R140Q podocin mRNA expression was elevated, protein abundance was reduced by more than 50% within one week following induction PMID: 29049388
  2. ATP evokes activation of TRPC6 channels in podocytes through P2Y receptors, podocin, and generation of reactive oxygen species. PMID: 24553432
  3. the carboxyl terminus of podocin/MEC-2 has to be placed at the inner leaflet of the plasma membrane to mediate cholesterol binding and contribute to ion channel activity. PMID: 24596097
  4. Podocin is necessary for assembly of the NOX2-TRPC6 complex in lipid rafts. PMID: 23948707
  5. study shows a variety of mechanical stimuli cause activation of endogenously expressed TRPC6 channels of podocytes; also that podocin reduces sensitivity of TRPC6 channels to mechanical stimuli, so that a loss of podocin function would result in hyperactivation of TRPC6 channels in foot processes PMID: 23657570
  6. These data descrived the importance of concomitant decreases in podocin, CD2AP and tensin2 during septic acute renal failure-associated proteinuria. PMID: 21799297
  7. we are for the first time able to prove the expression of a novel podocin isoform (isoform 2), exclusively and constitutively expressed in human podocytes, and reveal singular extrarenal podocin expression in human and murine testis PMID: 21499232
  8. Data show that the main slit diaphragm proteins, nephrin and podocin, are affected from the earlier stages of lupus nephritis and their expression correlates with disease histology. PMID: 21478284
  9. Coexpression of CIN85/Ruk(L) with CD2AP led to a decreased binding of CIN85/Ruk(L) to nephrin and podocin, which indicates a functional competition between CD2AP and CIN85/Ruk(L). PMID: 20457601
  10. Nphs2-/- mice developed proteinuria during the antenatal period and died a few days after birth from renal failure caused by massive mesangial sclerosis. PMID: 14701729
  11. The abnormality of a single podocyte molecule, caused by a single gene mutation in podocin can lead to hereditary nephrotic syndromes. PMID: 16501493
  12. Results suggest that nephrin Y1193 serves as a molecular switch that determines the integrity of the slit diaphragm by functional competition between beta-arrestin2 and podocin. PMID: 16968782
  13. MEC-2, Podocin, and probably many other PHB-domain proteins by binding to themselves, cholesterol, and target proteins regulate the formation and function of large protein-cholesterol supercomplexes in the plasma membrane PMID: 17079490
  14. Mutations in the NPHS2 gene, encoding podocin, are responsible for familial autosomal recessive and sporadic cases of steroid-resistant nephrotic syndrome PMID: 18288100
  15. Podocin inactivation in mature kidneys causes focal segmental glomerulosclerosis and nephrotic syndrome. PMID: 19713307
