Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Ligand-gated chloride channel which is a component of the heteropentameric receptor for GABA, the major inhibitory neurotransmitter in the brain. Plays an important role in the formation of functional inhibitory GABAergic synapses in addition to mediating synaptic inhibition as a GABA-gated ion channel. The gamma2 subunit is necessary but not sufficient for a rapid formation of active synaptic contacts and the synaptogenic effect of this subunit is influenced by the type of alpha and beta subunits present in the receptor pentamer. The alpha1/beta2/gamma2 receptor, alpha2/beta2/gamma2 receptor and the alpha1/beta3/gamma2 receptor exhibit synaptogenic activity whereas the alpha2/beta3/gamma2 receptor shows very little or no synaptogenic activity. Functions also as histamine receptor and mediates cellular responses to histamine.
Gene References into Functions
  1. RNF34 regulates postsynaptic gamma2-GABAAR clustering and GABAergic synaptic innervation by interacting with and ubiquitinating the gamma2-GABAAR subunit promoting GABAAR degradation PMID: 25193658
  2. This study demonistrated that Gabrg2 receptors are located in cholinergic terminals in the nucleus pontis oralis of the rat. PMID: 24141149
  3. This study provides evidence that alpha1F64, a key aromatic residue at the GABA binding site, is involved in both the binding and gating properties of alpha1beta1gamma2 receptors. PMID: 24573278
  4. Switching the intracellular domains of the gamma2 and gamma1 subunits substantiated this inference. Because this region determines post-synaptic localization, we hypothesize that GABAARs containing gamma1 and gamma2 use different mechanisms for synaptic clustering. PMID: 24425869
  5. These data demonstrate that altered expressions of GABA(A)R (alpha4, gamma2) and KCC2 and astrogliosis observed in TRMs hippocampus may provide us good therapeutic targets for the treatment of genetic epilepsy. PMID: 21924329
  6. Significant reduction of gamma2 protein was observed in the hippocampus of the post-traumatic epilepsy group compared to control. PMID: 21439793
  7. dispersal of GABA(A)R clusters and increased mobility of individual GABA(A)Rs are dependent on serine 327 within the intracellular loop of the GABA(A)R gamma2 subunit PMID: 20823221
  8. The gamma2 subunit short splice variant serves dual roles, as an integral subunit of the GABA(A) receptor and as an accessory protein acting as an external modulator of GABA(A) receptor function. PMID: 20371809
  9. The role of Gabrg2 in channel conductance in hippocampal neurons and the mechanism of protein interactions are reported. PMID: 19703932
  10. Elevated quantities of GABA(A) receptor gamma 2 subunit mRNA in the rat are related to the highly advanced behavioral and mnemonic strategies that are under the control of the telencephalon in this mammal. PMID: 11845246
  11. Prenatal DZ exposure produces functional modifications of the hippocampal GABA(A) receptor modulating NA release. DZ may affect gamma 2 subunit expression, so the receptor can be activated by muscimol but is not susceptible to allosteric modulation. PMID: 12145412
  12. during synaptic maturation GABAAR gamma2 and beta3 subunits are able to associate with synaptic and extrasynaptic clusters of gephyrin; data are compatible with a role of gephyrin in GABAAR synaptic stabilization rather than in clusters formation PMID: 12812758
  13. Transient kinetic techniques allow one to determine not only the channel-opening probability and rates of receptor desensitization but also the opening and closing rates of the mutated GABA(A) receptor channel PMID: 15182195
  14. the postsynaptic clustering of GABA(A)Rs in pyramidal neurons is involved in the stabilization of the presynaptic GABAergic contacts PMID: 16248887

  16. GABA-A receptor subunit gamma 2-arginine-43 domain is involved in the control of receptor assembly that may be relevant to the effect of the heterozygous gamma 2(R43Q) mutation leading to childhood absence epilepsy and febrile seizure. PMID: 17148443

  21. Data suggest that 3alpha,5alpha-THP's actions on dopamine type 1-like and/or GABAA receptors in the ventral tegmental area may require activity of protein kinase C. PMID: 18675324
  22. a likely explanation for the generalized epilepsy with febrile seizures plus phenotype is a dominant-negative suppression of wild-type receptors by the mutant gamma2S subunit in combination with loss of mutant gamma2S subunit protein function. PMID: 19261880
  23. Since the gamma2 subunit is involved with GABA(A) receptor trafficking and synaptic clustering, it appears that this subunit could be an important component of the activity-dependent response of the spinal cord after a spinal injury. PMID: 19358834
  24. Collectively, our findings suggest that GABA(A) but not GABA(B) receptors in the tLH act to suppress feeding behavior. PMID: 19501070
