The product CSB-E12649m is an in vitro ELISA kit for the quantitative measurement of Aspartate Aminotransferase (AST) in mouse serum, plasma, or cell culture supernates. AST is a pyridoxal phosphate (PLP)-dependent transaminase enzyme found in the liver, heart, skeletal muscle, kidneys, brain, and red blood cells. AST plays an important role in amino acid metabolism because, in the presence of PLP (Vitamin B6), it catalyzes the transfer of the amino group from aspartate or glutamate to the corresponding ketoacid. Serum AST level, serum ALT (alanine transaminase) level, and AST/ALT ratio are commonly measured clinically as biomarkers for liver health. So the AST test is used to check the liver damage.

This kit uses the quantitative sandwich-based enzyme immunoassay technique to measure the amount of mouse AST in the sample. Standards and samples are respectively added to the microplate wells pre-coated with an anti-mouse AST antibody. Biotin-labeled AST antibody, HRP-avidin, TMB substrate are pipped into the microplate in turn. The AST in the samples or standards is sandwiched between pre-coated AST antibody and Biotin-conjugated AST antibody, forming immune complexes. The color renders blue after the addition of TMB substrate. The addition of the stop solution into the wells immediately turns the blue into yellow. The concentration of AST in the samples is directly proportional to OD (450nm). Each manufactured lot of this ELISA kit was quality tested for criteria such as sensitivity, specificity, precision, linearity, and lot-to-lot consistency. Materials provided
  • A micro ELISA plate ---The 96-well plate has been pre-coated with an anti-Mouse AST antibody. This dismountable microplate can be divided into 12 x 8 strip plates.
  • Two vials lyophilized standard ---Dilute a bottle of the standard at dilution series, read the OD values, and then draw a standard curve.
  • One vial Biotin-labeled AST antibody (100 x concentrate) (120 μl/bottle) ---Act as the detection antibody.
  • One vial HRP-avidin (100 x concentrate) (120 μl/bottle) ---Bind to the detection antibody and react with the TMB substrate to make the solution chromogenic.
  • One vial Biotin-antibody Diluent (15 ml/bottle) ---Dilute the Biotin-antibody.
  • One vial HRP-avidin Diluent (15 ml/bottle) ---Dilute the HRP-avidin solution.
  • One vial Sample Diluent (50 ml/bottle)---Dilute the sample to an appropriate concentration.
  • One vial Wash Buffer (25 x concentrate) (20 ml/bottle) ---Wash away unbound or free substances.
  • One vial TMB Substrate (10 ml/bottle) ---Act as the chromogenic agent. TMB interacts with HRP, eliciting the solution turns blue.
  • One vial Stop Solution (10 ml/bottle) ---Stop the color reaction. The solution color immediately turns from blue to yellow.
  • Four Adhesive Strips (For 96 wells) --- Cover the microplate when incubation.
