Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw
cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature
and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized
form is 12 months at -20°C/-80°C.

Description
To produce the recombinant human hypoxanthine-guanine phosphoribosyltransferase (HPRT1) in an E. coli expression system, a series of steps are followed. Initially, the gene fragment encoding the specific region of the human HPRT1 protein (amino acids 2-218) is inserted into a suitable expression vector, along with an N-terminal 6xHis-tag. This recombinant construct is then introduced into E. coli cells through a transformation process. Following transformation, the cells are screened to identify positive clones, and a large-scale culture is initiated to facilitate efficient protein expression. The presence of the N-terminal 6xHis-tag simplifies the subsequent purification and detection of the recombinant HPRT1 protein. Upon cell lysis, the recombinant HPRT1 protein is isolated and purified from the cellular lysate. The purity of the recombinant HPRT1 protein is evaluated through SDS-PAGE analysis, indicating a high level of purity, up to 90%. When visualized on the gel, the purified recombinant HPRT1 protein appears as a well-defined band with an estimated molecular weight of approximately 33 kDa.
