Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
After binding acetylcholine, the AChR responds by an extensive change in conformation that affects all subunits and leads to opening of an ion-conducting channel across the plasma membrane.
Gene References into Functions
  1. Data suggest that the Chrna5 D398N variant influences the effect of developmental nicotine exposure on nicotine intake of exposed offspring. This gene-environment interaction on striatal DA release may provide motivation for increased nicotine seeking in N Nic offspring and reduced consumption in D Nic offspring. PMID: 29573323
  2. The findings reveal a proaddiction adaptive response to chronic nicotine in which nitric oxide and SST are released by a specific CHRNA5-expressing neuronal population to provide retrograde inhibition of the habenula- interpeduncular nucleus circuit and thereby enhance the motivational properties of nicotine. PMID: 29158387
  3. Genetic deletion of alpha5 nAChR subunits leads to a behavioural phenotype that exactly matches that observed after antagonizing dopamine receptors. PMID: 28498560
  4. Chrna5 KO mice did not exhibit high responses to novelty but showed decreased novelty-induced rearing behaviour together with high anxiety. Exposure to high doses of nicotine rescued these phenotypes. Results suggest that altered alpha5*-nAChR cholinergic signalling contributes to emotional/behavioural impairments that may be alleviated by nicotine consumption. PMID: 27385416
  5. Results suggest that alpha5-containing nAChRs mediate analgesic tolerance to nicotine but do not play a major role in neuropathic pain PMID: 25725336
  6. These results highlight the involvement of the alpha5 nAChR subunit in the development of hyperalgesia, allodynia and inflammation associated with chronic neuropathic and inflammatory pain models. PMID: 25931144
  7. the alpha5 subunit of the nicotinic acetylcholine receptor has a critical role in defining the sensitivity of the Ventral Tegmental Area Dopamine system to nicotine. PMID: 24296975
  8. Overexpression of the genomic cluster CHRNA5/A3/B4 in mice increases somatic signs associated with morphine withdrawal and Knockout of alpha5 gene reduced somatic signs in morphine-treated mice PMID: 24750073
  9. The alpha5 subunit is critical for controlling the expression and functional role of a population of alpha4*-containing nAChRs in the ventral-tegmental area. PMID: 23869214
  10. alpha5 in wild-type mice assembles into receptors that also contain the subunits alpha3, beta2, and beta4 PMID: 22380605
  11. alpha5 nicotinic receptor subunits are critical to the regulation of dopamine transmission by alpha4beta2 receptors in regions of dorsal striatum associated with habitual and instrumental responses. PMID: 22396410
  12. Normal developmental changes in nicotinic signaling and dendritic morphology in prefrontal cortex depend on alpha5-comprising nicotinic acetylcholine receptors. PMID: 22030359
  13. These findings suggested that disrupting nicotinic receptors fundamentally alters the mechanisms and timing of excitation in prefrontal attentional circuitry. PMID: 22072695
  14. Data reveal a highly regulated temporal-spatial pattern of alpha5 nAChR subunit expression during important periods of lung morphogenesis by FoxA2. PMID: 21682884
  15. increased nicotine intake in mice with a null mutation in Chrna5 PMID: 21278726
  16. This study demonstrated a novel and important role for the nicotinic receptor 5 subunit in adult brain circuitry required for attentional performance. PMID: 20610759
  17. functional role and pharmacological properties PMID: 12122136
  18. Alpha 5-containing nicotinic acetylcholine receptors participate in the modulation of colitis in mice PMID: 15973160
  19. beta2-containing nAChRs are involved in the affective signs of nicotine withdrawal, whereas non-beta2-containing nAChRs are more closely associated with physical signs of nicotine withdrawal PMID: 18184829
  20. Alpha5 nicotinic receptor gene deletion exerts a small but significant decrease in maximal acetylcholine-evoked [3H]gamma-aminobutyric acid release in hippocampus and striatum, with a more profound effect in cortex. PMID: 19139153
