Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
ATP-dependent RNA helicase required during spermatogenesis to repress transposable elements and prevent their mobilization, which is essential for germline integrity. Acts via the piRNA metabolic process, which mediates the repression of transposable elements during meiosis by forming complexes composed of piRNAs and Piwi proteins and governs the methylation and subsequent repression of transposons. Involved in the primary piRNA metabolic process. Specifically binds to piRNA precursors and promotes the generation of intermediate piRNA processing fragments that are subsequently loaded to Piwi proteins. Acts via its ATP-dependent RNA helicase activity: displays 5'-3' RNA unwinding activity and probably mediates unwinding and funneling of single-stranded piRNA precursor transcripts to the endonuclease that catalyzes the first cleavage step of piRNA processing to generate piRNA intermediate fragments that are subsequently loaded to Piwi proteins.; May act downstream of MEF2C during heart formation. Acts as a cardiac-specific suppressor of cardiomyocyte hypertrophy and cell cycle progression, suggesting that it may suppress these processes through the regulation of CDKN1A. Such results however require additional evidence.
Gene References into Functions
  1. Conditional inactivation of the putative RNA helicase Mov10l1 in mouse spermatocytes produces a specific loss of pachytene piRNAs, significant accumulation of pachytene piRNA precursor transcripts, and unusual polar conglomeration of Piwi proteins with mitochondria. Mov10l1 is required for pachytene piRNA biogenesis. PMID: 23166510
  2. Mutations in the conserved ATP hydrolysis motif of MOV10L1 block piRNA biogenesis and result in male sterility. PMID: 27655786
  3. MOV10L1 RNA helicase activity promotes unwinding and funneling of the single-stranded piRNA precursor transcripts to the endonuclease that catalyzes the first cleavage step of piRNA processing PMID: 25762440
  4. MOV10L1 exhibits 5'-to-3' directional RNA-unwinding activity in vitro and its RNA helicase activity is required for primary piRNA biogenesis in vivo. MOV10L1 selectively binds piRNA precursor transcripts. MOV10L1 is essential for the generation of intermediate piRNA processing fragments. piRNA precursor processing is intimately coupled with elements of local secondary structures such as G quadruplexes. PMID: 25762440
  5. MOV10L1 is essential for the primary processing of pachytene piRNAs. MOV10L1 acts upstream of PIWI proteins in the piRNA pathway and is a master regulator of the piRNA biogenesis. PMID: 23166510
  6. Mov10l1 mutant males are sterile owing to complete meiotic arrest. This mutant expresses Piwi proteins but lacks piRNAs, suggesting that MOV10L1 is required for piRNA biogenesis and/or loading to Piwi proteins. PMID: 20534472
  7. studies reveal a crucial role of MOV10L1 in male fertility and piRNA-directed retrotransposon silencing in male germ cells and suggest that MOV10L1 functions as a key component of a safeguard mechanism for the genetic information in male germ cells PMID: 20547853
  8. expression in oocytes sensitive to TAF4B level PMID: 19684329
  9. CHAMP acts as an inhibitor of cell proliferation and cardiomyocyte hypertrophy. PMID: 11854500
  10. Nkx2.5 transactivates the Csm promoter, suggesting that Nkx2.5 is essential for embryonic Csm (cardiac-specific isoform of Mov10l1) expression PMID: 12754203
