Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Sodium/ascorbate cotransporter. Mediates electrogenic uptake of vitamin C, with a stoichiometry of 2 Na(+) for each ascorbate.
Gene References into Functions
  1. SVCT2 was also detected in progenitor cells located in the inner and outer subventricular zone PMID: 27596508
  2. This study found impaired myelination and reduced collagen expression in Sodium-dependent Vitamin C Transporter 2 heterozygous mice (SVCT2(+/-) ) during peripheral nerve development and after peripheral nerve injury. PMID: 28456003
  3. Data show that phosphorylation of caveolin-1 on Tyr(14) in microglia induced the internalization of sodium-vitamin C cotransporter 2 (SVCT2). PMID: 28351945
  4. Study shows that SVCT2 is regulated at the post-transcriptional level by miR-141 and miR-200a during osteogenic differentiation of bone marrow stromal cells. PMID: 25617715
  5. Results indicate that both the SVCT2 transporter and oxidative stress play a vital role in BMSC attachment, migration and cytoskeletal re-arrangement. PMID: 24365600
  6. The functional expression of SVCT2 was detected in HEK293 cells. The kinetic analysis suggested that an ascorbate-dependent mechanism accounts for targeted SVCT2 expression in the developing kidney during medullary epithelial cell differentiation. PMID: 22990596
  7. Using in vitro studies, we suggest a molecular interaction between the SVCT2wt and SVCT2sh isoforms, which may regulate the affinity of vitamin C uptake PMID: 24947427
  8. Increased expression of SVCT2 in a new mouse model raises ascorbic acid in tissues and protects against oxidative stress. PMID: 22558179
  9. the expression and transport activity of SVCT2 in brain capillary endothelial cells after transient ischemia PMID: 21347255
  10. Expression of various collagen types is reduced in sciatic nerves of SVCT2(+/-) heterozygous mice compared to wild-type. PMID: 22114285
  11. The results suggest that expression of the SVCT2 is differentially regulated during embryonic development and in adulthood. PMID: 22001929
  12. Selective macrophage ascorbate deficiency suppresses early atherosclerosis. PMID: 20974251
  13. Evidence of increased apoptosis in SVCT2(-/-) mice and disruption of the basement membrane in fetal brain. PMID: 20541602
  14. These results provide evidence for the first time that uptake of ascorbic acid in the peripheral nervous system is crucially dependent on the expression and activity of SVCT2. PMID: 19672970
  15. an ascorbic acid transporter essential for vitamin c transport into the brain, and for perinatal survival PMID: 11984597
  16. A deficiency of Ca(2+) or PO(4)(3-) inhibits SVCT2 expression in a time-dependent manner. PMID: 12788230
  17. We examined the roles of L-ascorbic acid (AA) and its transporter, sodium-dependent vitamin C transporter (SVCT) 2, in the Zn-induced expression of osteoblastic differentiation markers. PMID: 14622981
  18. SVCT2 is highly expressed in the apical membranes of mouse hypothalamic tanycytes. Vitamin C uptake mechanisms present in these cells may perform a neuroprotective role concentrating vitamin C in this brain area . PMID: 15625716
  19. Our results revealed that maintenance of intracellular ascorbate as a result of SVCT2 activity is crucial for neuronal development, functional maturation, and antioxidant responses. PMID: 17304569
  20. PGE2-induced SVCT2 plasma membrane translocation through EP4 receptor and phosphorylation of SVCT2 by PKA increases ascorbic acid uptake and osteoblast-like differentiation in MC3T3-E1 cells. PMID: 17585336
  21. These results show that mature erythrocytes fail to concentrate ascorbate due to the loss of SVCT2 during maturation in the bone marrow. PMID: 17586466
  22. Transcription of the SVCT2 gene could be positively or negatively modulated by the presence of oxidant (H(2)O(2)) or antioxidant (lipoate) compounds, respectively. PMID: 17643393
  23. This study showed that sodium-dependent vitamin C transporter 2 mediated increases in ascorbic acid uptake by the adrenals followed by excessive production of plasma norepinephrine may play a pivotal role in the development of diabetic complications. PMID: 17689499
  24. all three proximal tubule segments expressed the transporter but the S3 segment had the highest expression; Ascorbic acid transport in these cells was regulated by a single kinetic component that depended on the sodium concentration, pH and temperature PMID: 18614995
  25. Culture of the macrophages with oxLDL enhanced ascorbate uptake. This was associated with increased expression of the ascorbate transporter (SVCT2), which was prevented by ascorbate and by inhibiting the NF-kappaB pathway PMID: 19254685
  26. knock-out mice die at birth due to tissue ascorbate deficiency PMID: 19361554
