ABCD1, the peroxisomal half-transporter whose dysfunction causes X-linked adrenoleukodystrophy, is notoriously difficult to produce as a full-length integral membrane protein, yet this recombinant human ABCD1 (aa 1–745) retains the conformational integrity needed for meaningful interaction studies. Functional ELISA confirms that immobilized ABCD1 binds its physiological peroxisomal docking partner PEX19 with an EC50 of 22.96–33.00 µg/ml, providing a validated foundation for ligand-binding assays, cargo-release experiments, and membrane transporter functional studies. The full-length construct, expressed in an *E. coli* cell-free system with a C-terminal 10×His tag, also supports structural and biophysical characterization, antibody development, and use as an ELISA standard. Purity exceeding 90% by SDS-PAGE aligns with standards expected in drug-protein binding studies and quantitative interaction assays where background interference must remain minimal.
