Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Plays an important role in the differentiation and development of pancreatic islet beta cells. Transcriptional repressor that competes with PAX6 in binding to a common element in the glucagon, insulin and somatostatin promoters.
Gene References into Functions
  1. Down-regulation of PAX4 or its target gene encoding the p97/VCP ATPase reduced myofibril disassembly. PMID: 28096335
  2. Ectopic expression of Pax4 in pancreatic delta cells results in beta-like cell neogenesis. PMID: 29025873
  3. The coordinated regulation of distinct cellular pathways particularly related to ER homeostasis by PAX4 not achieved by the mutant variant PAX4R129W alleviates beta cell degeneration and protects against diabetes mellitus. PMID: 26813254
  4. This work establishes the existence of a bona fide PAX4-enriched cell subpopulation nested within pancreatic islets that can expand in periods of increased functional demands, and exhibit improved viability in response to pathophysiological situations. PMID: 26503027
  5. Pax4 expression is not sufficient to transduce pancreatic alpha cells into beta cells. Overexpression of Pax4 in alpha cells may not increase functional beta cell number in diabetic patients PMID: 26183751
  6. Adult glucagon producing cells can be converted into beta-like cells upon Pax4 misexpression. PMID: 23810513
  7. these finding demonstrate that, downstream of Neurog3, the specification of a subset of enteroendocrine subtypes relies on both Arx and Pax4, while others depend only on Arx or Pax4. PMID: 22570716
  8. Pax4 protects adult islets from stress-induced apoptosis by suppressing selective nuclear factor-kappaB target genes while increasing Bcl-2 levels. PMID: 21521872
  9. Pax4 has a role in facilitating embryonic stem cell differentiation into the pancreatic lineage PMID: 12525695
  10. A2 and E1 elements in endocrine promoters are essential for Pax4 expression PMID: 14648838
  11. activity appears essential for appropriate initiation of beta-cell differentiation PMID: 14729487
  12. Pax4 is an essential regulator of pancreatic beta-cell development [review] PMID: 15650323
  13. Pax-4 and Arx act as transcriptional repressors that control the expression level of one another, thereby mediating proper endocrine fate allocation in the pancreas. PMID: 15930104
  14. The higher insulin production with glucose dependent modulation suggests that pax4 expression can drive more mature insulin producing cells. PMID: 17461469
  15. Data further support the notion that Pax4 activity is necessary to establish appropriate patterns of gene expression in endocrine progenitors of the digestive tract. PMID: 18058910
  16. PAX4 gene is not associated with the risk of type 1 diabetes in a Han Chinese sample. PMID: 18617287
  17. The daily changes in Pax4 expression may contribute to daily changes in function in the differentiated retinal photoreceptor. PMID: 19012751
  18. Ectopically expressed Pax4 using different cell-specific promoters demonstrate that Pax4 forces endocrine precursor cells, as well as mature alpha cells, to adopt a beta cell destiny. PMID: 19665969
