Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Terminates the action of GABA by its high affinity sodium-dependent reuptake into presynaptic terminals.
Gene References into Functions
  1. the expression of inhibitory neurotransmitter transporters GlyT1, GAT-1, and GAT-3 in inferior colliculus and hippocampus astrocytes, was examined. PMID: 29370841
  2. Results demonstrated that GAT-1-/- mice have phenotypes of hyperactivity, impaired sustained attention and learning deficiency, and the performance of GAT-1-/- mice is similar to ADHD symptoms. PMID: 26072958
  3. Results demonstrate that high-affinity activation by GABA of GAT1 transporters provokes release of [3H]GABA from nerve terminals by different mechanisms PMID: 25150942
  4. these data show that during paclitaxel-induced neuropathic pain there is significant increase in GAT-1 expression in the ACC. GAT-1 is the main transporter of GABA from the synapse PMID: 25700370
  5. Development of GABA-removal system and its relationship to development of GABAergic networks studied by examination of changes in GAT-1 and GAT-3 expression and localization in embryonic and postnatal mouse spinal cord PMID: 24412234
  6. GAT1 plays a critical role in the regulation of rapid eye movement (REM) sleep and homeostasis of non-REM sleep. PMID: 24155871
  7. results suggest that GAT1 ko mouse is a new animal model for ADHD studying and GAT1 may be a new target to treat ADHD PMID: 23656791
  8. We failed to provide proof-of-concept evidence that lower PV and GAT1 expression in schizophrenia are a consequence of lower GAD67 expression PMID: 23103418
  9. Results suggest that a moderate reduction in GAT1 activity causes the enhancement of learning and memory in mice. PMID: 22318715
  10. We used immunohistochemistry to analyze the spatiotemporal patterns of GABA and GABA Transporter-1 (GAT1) immunoreactivities in the developing and adult mouse amygdala PMID: 21382352
  11. the terminal sialic acid of N-linked oligosaccharides of GAT1 plays a crucial role in the gamma aminobutyric acid transport process PMID: 21045010
  12. GABA Transporter Mutagenesis Database (GATMD), a web-accessible, relational database of manually annotated biochemical, functional and pharmacological data reported on GAT1. PMID: 21131297
  13. abnormal expression of GAT1 could result in spermiogenesis function injury, sperm paramorphia and dysgenesis. PMID: 19938951
  14. GAT1 deletion reduces the frequency of hippocampal theta oscillation in vivo; behavioral studies show that GAT1 knock-out mice exhibit impaired hippocampus-dependent learning and memory. PMID: 20016099
  15. GAT1 deficiency leads to enhanced extracellular GABA levels resulting in an overactivation of hippocampal GABA-A receptors responsible for a postsynaptic tonic conductance. PMID: 12815026
  16. The rewarding effects induced by morphine were significantly decreased in GAT1-overexpressing mice as measured by the conditioned place preference (CPP) paradigm. Morphine withdrawal symptoms were substantially reduced in GAT1-overexpressing mice. PMID: 14598306
  17. GAT1 abnormal expression may result in gonadal dysgenesis and might be therapeutically targeted for contraception or dysgenesis treatment. PMID: 15040890
  18. It is concluded that GABA GAT-1 transporters and glutamate transporters coexist on the same spinal cord glutamatergic terminals. Activation of these GABA transporters elicits release of glutamate partially by reversal of glutamate transporters. PMID: 15789377
  19. results identify the transport mode and the channel mode of GAT1 and also raise the possibility of separating these components in a physiological environment PMID: 16150932
  20. Increased GABA transporters is associated with Niemann-Pick disease, type C PMID: 16429462

  26. In the developing murine postnatal neostriatum GAT-1 operates in a net uptake mode. It prevents the persistent activation of presynaptic GABA(B)Rs (P12-34) and prevents (P7-14) or reduces (P19-34) tonic postsynaptic GABA(A)R activity. PMID: 18832421
  27. These findings demonstrate that GABA uptake and release via GAT-1, GAT-3, and VGAT are initially established in the inner retina during the first postnatal week and that these systems then mature in the outer retina during the second postnatal week. PMID: 18975268
