CXCL6 (GCP-2) is a potent ELR+ CXC chemokine that signals through CXCR1 and CXCR2 to drive neutrophil recruitment, making it a critical tool for dissecting innate immune trafficking mechanisms. This tag-free recombinant human CXCL6, spanning residues 43–114 of the mature processed sequence, demonstrates robust chemotactic activity on primary human neutrophils at 10–50 ng/ml, providing a suitable basis for Transwell migration assays, CXCR1/CXCR2 signaling pathway studies involving MAPK and NF-κB activation, and in vivo neutrophilic inflammation models. Endotoxin levels below 1.0 EU/μg minimize LPS-driven artifacts that can confound neutrophil priming and cytokine release readouts, which satisfies the stringent criteria typical for primary immune cell functional assays. Produced in *E. coli* with greater than 98% purity by SDS-PAGE, this recombinant CXCL6 protein also supports use as a reference standard in chemokine detection assays and as a coating antigen for anti-CXCL6 antibody validation by ELISA.
