Regarding your ELISA kit CSB-E06799p, could you please advise on the sample types that you lab has tested with this kit, and what the observed concentration values were?
I was hoping to use their porcine cartilage homogenate samples with the kit. I have already homogenized the samples in RIPA buffer, and I was hoping you might be able to advise whether or not you thought RIPA might be a suitable lysis buffer for my samples?

We tested undiluted serum and plasma samples before and the test value is ND--30ng/ml. Pls find attached QC.
For cartilage samples, sorry we didn't use RIPA buffer before. Usually we use pretreatment combined with repeated freezing and thawing method.
Generally, RIPA will contain components such as SDS, Triton X-100, and NP-40. Pls note:
1. It must not contain SDS (which will destroy the hydrogen bond of protein macromolecules causing protein denaturation);
2. For Triton X-100, NP 40 and other mild detergents, you should control the low concentration range (generally no more than 1%, but it not verified, so we are not clear about the specific impact);
3.EGTA and EDTA can be used as chelating agents for metal ions to remove metal ions, which has a great influence on BCA method, but we are not clear about the impact on elisa;
4 The sample should not contain NaN3 components, which will inhibit HRP activity and cause false negative.
Therefore, if your sample has been prepared, you can do a pretest first but we are not clear about the final test result; if not prepared, it is recommended to follow the recommended method in our manual.

Metalloproteinase inhibitor that functions by forming one to one complexes with target metalloproteinases, such as collagenases, and irreversibly inactivates them by binding to their catalytic zinc cofactor. Acts on MMP1, MMP2, MMP3, MMP7, MMP8, MMP9, MMP10, MMP11, MMP12, MMP13 and MMP16. Does not act on MMP14. Also functions as a growth factor that regulates cell differentiation, migration and cell death and activates cellular signaling cascades via CD63 and ITGB1. Plays a role in integrin signaling.
