ACKR2 (D6) functions as a decoy receptor that scavenges inflammatory CC chemokines without triggering classical signaling, making it a critical target for studying chemokine gradient regulation in inflammation and immunity. This full-length recombinant human ACKR2 (aa 1–384) is produced using a cell-free *E. coli* expression system, a strategy well suited for generating full-length seven-transmembrane GPCRs that are difficult to express in conventional cellular hosts, and it carries an N-terminal 10×His tag for oriented immobilization. Functional ELISA confirms ligand-binding activity, with immobilized ACKR2 at 1 μg/ml binding human CCL2 at an EC₅₀ of 23.52–30.99 μg/ml, providing a suitable basis for receptor-ligand interaction assays, competitive inhibition studies, and blocking antibody screening. Purity exceeding 85% by SDS-PAGE supports use as a positive control in binding assays and aligns with standards expected in therapeutic antibody epitope mapping and small-molecule inhibitor screening campaigns.
