To assess the linearity of the assay, samples were spiked with high concentrations of human PDGF-BB in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.
The recovery of human PDGF-BB spiked to levels throughout the range of the assay in various matrices was evaluated. Samples were diluted prior to assay as directed in the Sample Preparation section.
These standard curves are provided for demonstration only. A standard curve should be generated for each set of samples assayed.
The human PDGF-BB ELISA Kit is engineered for accurate measurement of human PDGF-BB levels from samples including serum, plasma, or tissue homogenates. It uses the Sandwich-ELISA mechanism in combination with the enzyme-substrate chromogenic reaction to measure the PDGF-BB content in the sample. The color intensity is positively correlated with PDGF-BB content in the sample. The PDGF-BB concentration can be calculated according to the standard curve. This kit is tested with high sensitivity, strong specificity, good linearity, high precision and recovery, as well as lot-to-lot consistency.
PDGF-BB is a powerful promoter of cell proliferation and increases the expression of stem cell markers. It suppresses collagen production and mineralization but accelerates the maturation of collagen chains through increased LOX activity and SPARC expression. PDGF-BB binds to the PDGFR-β and subsequently activates several intracellular signaling cascades, including MAPK and PI3K/Akt pathways, which in turn activate the downstream targets mTOR and p70S6K. PDGF-BB-PDGFR-β signaling in brain pericytes is critical to the development, maintenance, and function of a healthy blood-brain barrier (BBB). It is also involved in the tissue repair and tumor progression effects and is released in response to pathogen-induced changes in the microenvironment acting as a rapid and early effector cytokine.
