Materials provided
  • A 96-well Assay plate--The 96-well plate has been pre-coated with goat-anti-rabbit ARO antibody.
  • Standard (6 x 0.5 ml)--Dilute the standard at dilution series, read the OD values, and then draw a standard curve.
  • HRP-conjugated ARO (1 x 6 ml) --Bind to the anti-fish ARO antibody, and HRP catalyzes the TMB to elicit a chromogenic reaction.
  • Anti-fish ARO Antibody (1 x 6 ml)--React with both ARO in the samples/standards and HRP-conjugated ARO.
  • Wash Buffer (20x concentrate) (1 x 15ml) --Wash away unbound or free substances.
  • Substrate A (1 x 7 ml) --Mix with Substrate B and interacts with HRP, eliciting the solution turns blue.
  • Substrate B (1 x 7 ml) --Mix with Substrate A and interacts with HRP, eliciting the solution turns blue.
  • Stop Solution (1 x 7ml) --Stop the color reaction. The solution color immediately turns from blue to yellow.

Description

The Fish Aromatase (ARO) ELISA Kit is suitable for the quantitative detection of ARO in fish samples, including serum, plasma, and tissue homogenates. It has been validated with excellent sensitivity, high specificity, premium precision, high recovery, and lot-to-lot consistency. See the product instructions for more details.

This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate has been pre-coated with goat-anti-rabbit ARO antibody. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for ARO and HRP-conjugated ARO. The competitive inhibition reaction is launched between with HRP-labeled ARO and ARO in the samples or standards with the anti-fish ARO antibody. The substrate solution is added to the wells and the color develops in opposite to the amount of ARO in the sample. The color development is stopped and the intensity of the color is measured.

ARO is the enzyme responsible for the aromatization of androgens to estrogen in many tissues, such as gonads, adipocytes, and the brain. It is important for sexual development. ARO helps make the estrogen that is conducive to fuel the growth of hormone-receptor-positive breast cancer. It may play a role in sex-determination. High levels of ARO was expressed at temperatures that yield female progeny in temperature-dependent sex determinant species. ARO also exerts a neuroprotective role. Many factors, including age, obesity, insulin, alcohol, and smoking, can enhance the activity of ARO.

The plate is coated with rabbit polyclonal antibody.
Immunogen is 293 cell-derived recombinant protein. It is affinity purified.
