Precision
Intra-assay Precision (Precision within an assay): CV%<8%
Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision (Precision between assays): CV%<10%
Three samples of known concentration were tested in twenty assays to assess.
Linearity
To assess the linearity of the assay, samples were spiked with high concentrations of Human ON in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

Description

This human osteonectin (ON) ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of human ON in different samples, including serum, plasma, saliva, or tissue homogenates. The enzyme-substrate chromogenic reaction is also used to amplify the signal and quantify the levels of the analyte through the intensity of the colored product. The color intensity positively correlates with the amount of ON bound in the initial step.

ON, also called SPARC, is encoded by the gene SPOCK2. It is a non-collagenous calcium-binding protein of bone matrix expressed in mineralized tissues including endothelial cells and fibroblasts. ON is involved in the adhesion of osteoblasts and platelets to their extracellular matrix and in early stromal mineralization. ON exerts these effects by binding strongly to type I collagen and hydroxyapatite. It also inhibits cell proliferation through the arrest of cells in the G1 phase of the cell cycle. ON expression is frequently increased in tissue wound repair or fibrotic diseases.

Target Background

Function
(From Uniprot)
May participate in diverse steps of neurogenesis. Binds calcium.
Gene References into Functions
  1. candidate susceptibility gene for bronchopulmonary dysplasia; lung expression pattern indicates potential role in alveolarization PMID: 21836138
  2. Neurons are a major source of all of the testican family members in the normal brain. Abundant distribution of testican 2 may contribute to glioma invasion by inactivating other testican family members including N-Tes, which all inhibit MT-MMPs. PMID: 12810672
  3. purification and characterization of a recombinant testican-2 expressed in insect cells; the C1q subcomponent of complement component C1 was identified as a potential interacting partner of testican-2 PMID: 17950619
Subcellular Location
Secreted, extracellular space, extracellular matrix.
Tissue Specificity
Highly expressed in brain. Also found in lung and testis.
