Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Description

IL-17F is a key mediator of mucosal immunity and chronic inflammatory signaling, driving neutrophil recruitment and pro-inflammatory gene expression through NF-κB and MAPK pathway activation. This tag-free recombinant human IL-17F, encompassing the full mature sequence (residues 31–163), demonstrates robust signaling potency with an ED50 below 20 ng/mL in an IL-6 secretion assay using murine NIH/3T3 cells, corresponding to a specific activity exceeding 5.0 × 10⁴ IU/mg — providing a reliable basis for cell-based functional assays, NF-κB/MAPK signaling studies, and use as a reference standard in cytokine detection platforms. IL17A-IL17F signals via IL17RA-IL17RC heterodimeric receptor complex, triggering homotypic interaction of IL17RA and IL17RC chains with TRAF3IP2 adapter through SEFIR domains. This leads to downstream TRAF6-mediated activation of NF-kappa-B and MAPkinase pathways ultimately resulting in transcriptional activation of cytokines, chemokines, antimicrobial peptides and matrix metalloproteinases, with potential strong immune inflammation. IL17A-IL17F is primarily involved in host defense against extracellular bacteria and fungi by inducing neutrophilic inflammation. As signature effector cytokine of T-helper 17 cells (Th17), primarily induces neutrophil activation and recruitment at infection and inflammatory sites. Stimulates the production of antimicrobial beta-defensins DEFB1, DEFB103A, and DEFB104A by mucosal epithelial cells, limiting the entry of microbes through the epithelial barriers.
