Description

This recombinant monoclonal antibody is designed as a research-grade biosimilar to TRK-950, targeting CAPRIN1 (Cytoplasmic Activation/Proliferation-Associated Protein 1), an RNA-binding protein that plays crucial roles in cellular stress responses and translational regulation. CAPRIN1 functions as a key component of stress granules, cytoplasmic aggregates that form during cellular stress to regulate mRNA translation and stability. This protein is involved in cell cycle progression, proliferation, and survival pathways. CAPRIN1 has been implicated in various pathological conditions, including cancer development and progression, where its overexpression correlates with enhanced tumor cell proliferation and metastatic potential. Additionally, CAPRIN1 participates in neuronal function and has been associated with neurodegenerative diseases and viral infections.

As a biosimilar to the reference antibody TRK-950, this recombinant monoclonal antibody provides researchers with a reliable tool for investigating CAPRIN1 expression, localization, and function in various cellular contexts. It enables studies examining stress granule dynamics, RNA metabolism, cancer biology, and cellular stress responses, supporting advancement in understanding CAPRIN1's role in both physiological and disease states.

Usage
It is a non-therapeutic biosimilar antibody, owning the same variable region from the corresponding approved therapeutic antibody. In conclusion, it is a research-grade biosimilar antibody and expressed in mammalian cell, which can be directly used as positive controls in drug discovery or used for rapid verification of the biological functions of target protein.

Target Background

Function
May regulate the transport and translation of mRNAs of proteins involved in synaptic plasticity in neurons and cell proliferation and migration in multiple cell types. Binds directly and selectively to MYC and CCND2 RNAs. In neuronal cells, directly binds to several mRNAs associated with RNA granules, including BDNF, CAMK2A, CREB1, MAP2, NTRK2 mRNAs, as well as to GRIN1 and KPNB1 mRNAs, but not to rRNAs.
Gene References into Functions
  1. that high Caprin1 expression was significantly associated with worse overall survival for patients with hepatocellular carcinoma PMID: 29037839
  2. Based on insights from the structures and existing biochemical data, the existence of an evolutionarily conserved ribonucleoprotein (RNP) complex consisting of Caprin-1, FMRP and G3BP1 is proposed. PMID: 27303792
  3. Data suggest that DEAD-box helicase 3 (DDX3X) physically interacts and co-localizes with poly(A)-binding cytoplasmic protein 1 (PABPC1) and caprin-1 in lamellipodia at the leading edge of spreading cells; these interactions are dependent on mRNA; depletion of DDX3X (via gene silencing with the CRISPR-Cas system) leads to decreased cell motility. These studies were conducted using MRC5 lung fibroblast cell line. PMID: 28733330
  4. G3BP mediates the condensation of stress granules by shifting between two different states that are controlled by the phosphorylation of S149 and by binding to Caprin1 or USP10. PMID: 27022092
  5. The G3BP1-Caprin1-PKR complex represents a new mode of PKR activation and is important for antiviral activity of G3BP1 and PKR during infection with mengovirus. PMID: 25784705
  6. Data show that tylophorine compounds exert anti-cancer activity predominantly by targeting and sequestering the caprin-1 protein and c-Myc mRNA associated ribonucleoprotein complex. PMID: 25669982
  7. G3BP1, G3BP2 and CAPRIN1 are required for translation of interferon stimulated mRNAs and are targeted by a dengue virus non-coding RNA. PMID: 24992036

  13. Suppression of expression of human Caprin-1 results in slowing of the proliferation rate, due to prolongation of the G1 phase of the cell cycle, formally demonstrating that Caprin-1 is essential for normal cellular proliferation. PMID: 16177067
  14. Caprin-1/G3BP-1 complex is likely to regulate the transport and translation of mRNAs of proteins involved with synaptic plasticity in neurons PMID: 17210633
