Target Background

Function
Seems to act as scaffold molecule at synaptic junctions by assembling neurotransmitter receptors and cell adhesion proteins. Plays a role in nerve growth factor (NGF)-induced recruitment of RAPGEF2 to late endosomes and neurite outgrowth. May play a role in regulating activin-mediated signaling in neuronal cells. Enhances the ability of PTEN to suppress AKT1 activation.
Gene References into Functions
  1. In summary,MAGI-2 and Fyn protect dendrin fromNedd4-2-mediated ubiquitination and from nuclear translocation, thereby maintaining the physiologic homeostasis of podocytes, and the lack of MAGI-2 in podocytes results in FSGS. PMID: 28539383
  2. Magi2 is an essential component of the slit diaphragm complex that is required for nephrin localization and foot process formation. PMID: 25993318
  3. MAGI-2 has a critical role in maintaining the functional structure of the slit diaphragm and this molecule has an essential role in the functioning of the kidney filtration barrier. PMID: 25108225
  4. We showed the expression patterns of MAGI-2 in detail in adult mice. Significant Venus expression was detected in various regions of the brain, also detected in glomeruli of the kidney. We found the novel expression of MAGI-2 in the testis. PMID: 22361463
  5. MAGI-2 is required for the integrity of the kidney filter and podocyte survival. PMID: 25271328
  6. elevation of S-SCAM expression in the excitatory neurons of forebrain was sufficient to induce an schizophrenia-like phenotype in mice. PMID: 25653350
  7. In Usher syndrome 1G, mutations in SANS eliminate Magi2 binding and thereby deregulate endocytosis, lead to defective ciliary transport modules and ultimately disrupt photoreceptor cell function inducing retinal degeneration. PMID: 24608321
  8. ARIP1 and ARIP2 are co-expressed in some nerve cells and their biological activities are distinct. PMID: 23523645
  9. Two subsynaptic domains, one containing IgSF9b for synaptic adhesion and the other containing gephyrin and GABAA receptors for synaptic transmission, are interconnected through S-SCAM and neuroligin 2. PMID: 23751499
  10. ARIP1 is a key molecule for the regulation of the action of activin in neurones, and also that decreased ARIP1 expression in the lesioned brain may be beneficial to the neurotrophic and neuroprotective roles of activin A in recovery after brain injury. PMID: 22849377
  11. Localization of synaptic scaffolding molecule (S-SCAM) at the puncta adherentia junctions formed between the mossy fibre terminals and the dendrites of CA3 pyramidal cells. PMID: 14750953
  12. These experiments identify MAGI-2 as a strong candidate for linking TARP/AMPA receptor complexes to a wide range of other postsynaptic molecules and pathways and advance our knowledge of protein interactions at mammalian CNS synapses. PMID: 16870733
