Target Background

Function
Required for transport of secretory proteins from the Golgi complex. Catalyzes the transfer of phosphatidylinositol (PI) and phosphatidylcholine (PC) between membranes in vitro. Essential for viability and secretion. Exchanges its bound phospholipid with phospholipid monomers that reside in membrane bilayers. Regulates specific trans-Golgi export pathways, like transport from endosomes to the trans-Golgi or transport from the plasma membrane to the vacuole at the level of the endosome. Increased membrane curvature and lipid unsaturation levels at the trans-Golgi membrane promotes membrane binding and phospholipid transfer of SEC14. Thus, SEC14 may act at the trans-Golgi membrane to exchange lipids and promote vesicle formation. The phosphatidylcholine-bound form of SEC14 represses the CDP-choline pathway activity by inhibiting CCTase, the rate-determining enzyme of the CDP-choline pathway. PI binding/transfer is dispensable for function in vivo. Required for trafficking and localization of lipid raft proteins to the plasma membrane.
Gene References into Functions
  1. Data suggest that SEC14L and SFH1 undergo conformational changes as phosphatidylinositol/phosphatidylcholine bind to hydrophobic pockets/ligand binding sites. [review-like article] PMID: 25233419
  2. Sec14 and Sec14 like proteins form central conduits to integrate multiple aspects of lipid metabolism with productive phosphoinositide signaling. (Review) PMID: 22374094
  3. The crystal structure of Sec14 family homologue (Sfh)3 is the first report of a dimeric Sec14 protein and provides clues for its functional study. PMID: 23519406
  4. Localization of lipid raft proteins to the plasma membrane is a major function of the phospholipid transfer protein Sec14. PMID: 23383173
  5. To launch a detailed structural and functional characterization of Sfh3, the recombinant protein was purified to homogeneity, diffraction-quality crystals were produced and a native X-ray data set was collected to 2.2 A resolution PMID: 22102037
  6. Sec14 protein is a peripheral Golgi membrane protein essential for Golgi secretory function and cell viability[review]. PMID: 16118014

  8. The application of EPR spectrscopy to investigate the chemical properties of binding between phosphatidylcholine and Sec14 is reported. PMID: 16997918
  9. molecular dynamics suggests large rigid body motions within Sec14p accompany closing and opening of an A(10)/T(4)/A(11) helical gate, and that "state-of-closure" of this helical gate determines access to the Sec14p phospholipid binding cavity PMID: 17344474
