Target Background

Function
Required for rescue of stalled ribosomes mediated by trans-translation. Binds to tmRNA RNA (also known as SsrA or 10Sa RNA, 363 nucleotides in this organism), required for stable binding of tmRNA to ribosomes. tmRNA and SmpB together mimic tRNA shape, replacing the anticodon stem-loop with SmpB (Probable). tmRNA is encoded by the ssrA gene; the 2 termini fold to resemble tRNA(Ala) and it encodes a 'tag peptide', a short internal open reading frame. Able to recruit charged tmRNA to ribosomes. Does not play a role in transcription, processing or Ala-aminoacylation of tmRNA. Other studies have shown it stimulates aminoacylation of tmRNA. May protect tmRNA from degradation. Binds to tmRNA that cannot be aminoacylated (tmRNA G3A), does not bind to tmRNA mutations near the tRNA-like termini (tmRNA G19C, A334U); other tmRNA mutations that block trans-translation still bind SmpB. With tmRNA may play a role in bacterial persistence. During trans-translation Ala-aminoacylated transfer-messenger RNA acts like a tRNA, entering the A-site of stalled ribosomes, displacing the stalled mRNA. The ribosome then switches to translate the ORF on the tmRNA, the nascent peptide is terminated with the 'tag peptide' encoded by the tmRNA and targeted for degradation. The ribosome is freed to recommence translation, which seems to be the essential function of trans-translation.

  1. These data indicate that the tmRNA-SmpB system is important in the pathogenesis of Avian pathogenic E. coli O2 strain E058. PMID: 24013628
  2. Transfer-messenger RNA-SmpB protein regulates ribonuclease R turnover by promoting binding of HslUV and Lon proteases PMID: 22879590
  3. cryo-electron microscopy reconstruction of a fusidic-acid-stalled ribosomal 70S-tmRNA-SmpB-EF-G complex (carrying both of the large ligands, that is, EF-G and tmRNA) at 8.3 A resolution PMID: 22622583
  4. There is tight and specific binding of SmpB to transfer messenger (tm)RNA. Surprisingly, the global conformation and flexibility of tmRNA do not change upon SmpB binding, as a function of magnesium ion (Mg2+) concentration. PMID: 22482838
  5. Mutations in the SmpB tail reduce rates of peptidyl transfer to tmRNA, but not GTP hydrolysis by EF-Tu. PMID: 21795410
  6. Data conclude that the tmRNA.SmpB complex accommodates in the ribosomal A site very much like an aminoacyl-tRNA during protein elongation. PMID: 19883769
  7. truncation of the C-terminal tail or substitution of tryptophan residue at 147 in the middle of the C-terminal tail affected the activity in the early stage of trans-translation PMID: 20348441
  8. protein SmpB enhanced transfer-messenger RNA (tmRNA) processing, suggesting a new role for SmpB in trans-translation PMID: 15494393
  9. SmpB remains tmRNA bound at the initial steps of trans-translation PMID: 17400816
  10. These results suggest that SmpB serves to bridge two separate domains of tmRNA to determine the initial codon for tag-translation. PMID: 17698641
  11. within a stalled ribosome, SmpB interacts with the three universally conserved bases G530, A1492 and A1493 that form the 30S subunit decoding centre, in which canonical codon-anticodon pairing occurs PMID: 19132006
  12. These results demonstrate that SmpB is functionally linked with the sequence upstream of the tmRNA template; both contribute to reading frame selection on tmRNA. PMID: 19540849
