Target Background

Function
Transcription factor involved in the regulation of gene expression in response to extracellular amino acid levels. Synthesized as latent cytoplasmic precursor, which, upon a signal initiated by the plasma membrane SPS (SSY1-PTR3-SSY5) amino acid sensor system, becomes proteolytically activated and relocates to the nucleus, where it induces the expression of SPS-sensor-regulated genes, including the amino-acid permeases AGP1, BAP2, BAP3 and GNP1. Binding to promoters is facilitated by DAL81. Involved in the repression of genes subject to nitrogen catabolite repression and genes involved in stress response. Negatively regulated by inner nuclear membrane proteins ASI1, ASI2 and ASI3, which prevent unprocessed precursor forms that escape cytoplasmic anchoring from inducing SPS-sensor-regulated genes. May be involved in pre-tRNA splicing.
Gene References into Functions
  1. Tup1p bound to the promoter region of TAT1 and TAT2 genes that were dependent on STP1 and STP2 (encoding DNA-binding activator proteins) for expression. PMID: 26546823
  2. A novel class of STP1 mutations affecting RI were isolated that are less efficiently retained in the cytoplasm but remain under tight negative control by the Asi proteins. PMID: 25253722
  3. Data show that the Ssy5 prodomain is a potent inhibitor of chymotrypsin-like catalytic cleavage of Stp1 and that inactivation of Ssy5 is a requisite for receptor-activated proteolysis. PMID: 20421414
  4. post-whole genome duplication co-conservation of the STP1 and STP2 genes was favored by functional divergence of their products, likely conferring to cells an increased ability to adapt to various amino acid supply conditions. PMID: 19906648
  5. Stp1 may act in combination with distinct sets of transcription factors, according to the gene context, to promote induction of transcription in response to external amino acids. PMID: 15126393
  6. there appears to be only one pathway for induction with L-citrulline, and this pathway is completely dependent on the SPS component, Ssy1p, and either of the transcription factors, Stp1p and Stp2p. PMID: 15197729
  7. Ssy5 is the endoprotease responsible for Stp1 processing PMID: 15509782
  8. The phosphatase complex containing Rts1p keeps the SPS-mediated pathway down-regulated in the absence of extracellular amino acids by dephosphorylating a component of the pathway. PMID: 16400180
  9. Asi1, Asi2, and Asi3 work together to maintain the latent properties of transcription factors Stp1 and Stp2 PMID: 17085444
  10. Dal81 indiscriminately amplifies the transactivation potential of both full-length and processed Stp1 and Stp2 by facilitating promoter binding. PMID: 17603098
  11. study reports that degradation of Stp1, a transcription factor for amino acid uptake and a key effector of the SPS amino-acid-sensing pathway, is controlled by TOR activity PMID: 19494127

Subcellular Location
Cell membrane; Peripheral membrane protein; Cytoplasmic side. Nucleus. Note=Localizes to the cytoplasm in its unprocessed form and is targeted to the nucleus after proteolytic processing upon induction by amino acids. The SCF(MET30) ubiquitin ligase complex negatively regulates nuclear accumulation of the processed form in the absence of high extracellular methionine levels.
