Target Background

Function
Releases the supercoiling and torsional tension of DNA introduced during the DNA replication and transcription by transiently cleaving and rejoining one strand of the DNA duplex. Introduces a single-strand break via transesterification at a target site in duplex DNA. The scissile phosphodiester is attacked by the catalytic tyrosine of the enzyme, resulting in the formation of a DNA-(5'-phosphotyrosyl)-enzyme intermediate and the expulsion of a 3'-OH DNA strand. The free DNA strand than undergoes passage around the unbroken strand thus removing DNA supercoils. Finally, in the religation step, the DNA 3'-OH attacks the covalent intermediate to expel the active-site tyrosine and restore the DNA phosphodiester backbone. Essential for proper chromosome segregation in both meiosis and mitosis. Weakly relaxes negative supercoils and displays a distinct preference for binding single-stranded DNA. The TOP3-SGS1 protein complex may function as a eukaryotic reverse gyrase introducing positive supercoils into extrachromosomal ribosomal DNA rings.
Gene References into Functions
  1. Although Top3 and Sgs1 interact without sumoylation, sumoylated forms of Sgs1, particularly those containing K621 conjugates, are preferentially enriched in complexes with Top3. PMID: 27373152
  2. Our data are consistent with a redundant role of Top2 and Sgs1 together with Top3 (Sgs1-Top3) in replication fork merging at rDNA barriers. PMID: 26630413
  3. Top3-Rmi1 acts in all meiotic recombination functions previously associated with Sgs1, most notably as an early recombination intermediate chaperone, promoting regulated crossover and noncrossover recombination. PMID: 25699707
  4. Top3 specifically disrupts D loops mediated by yeast Rad51/Rad54. PMID: 25699708
  5. The strand-passage activity of Top3-Rmi1 is required for all known functions of Sgs1 in meiotic recombination, including channeling JMs into physiological crossover and noncrossover pathways, and suppression of non-allelic recombination. PMID: 25699709
  6. the roles played by Top3 and the STR complex as a whole during the resolution of replication-associated recombination intermediates. PMID: 24100144
  7. Rmi1 has a unique regulatory capacity that slows DNA relaxation by Top3 but stimulates DNA decatenation. PMID: 22885009
  8. HJ resolvases and Sgs1-Top3-Rmi1 comprise two independent processes to deal with HJ-containing DNA intermediates arising during HRR in S-phase. PMID: 21383164
  9. Study established that Dna2, Sgs1 and RPA constitute a minimal protein complex capable of DNA resection in vitro; in addition it was found that both the Top3 and Rmi1 complex and the Mre11-Rad50-Xrs2 complex have important roles as stimulatory components. PMID: 20811461
  10. Loss of the TOP3 gene, encoding topoisomerase III, results in a phenotype of slow growth PMID: 15654096
  11. Rmi1 physically interacts with Sgs1 and Top3 and is a third member of this complex. PMID: 15889139
  12. The effects of TOP3 on telomere binding in human and S. cerevisiae cells are reported. PMID: 16546998
  13. The pif1 top3 mutant combination is synthetic lethal; Pif1-Top3 genetic interaction is recombination dependent. PMID: 16816432
  14. We propose that Top3 functions in S phase to both process homologous recombination intermediates and modulate checkpoint activity. PMID: 16899506
  15. A model is proposed in which the Shu proteins act in homologous recombination repair(HRR) to promote the formation of HRR intermediates that are processed by the Sgs1-Rmi1-Top3 complex. PMID: 17671161
  16. Top3-Rmi1 functions as a complex and suggest that Rmi1 stimulates Top3 by promoting its interaction with ssDNA. PMID: 17693398
  17. Mus81.Mms4 together with Rad54 efficiently process perturbed replication forks to promote recovery and may constitute an alternative mechanism to the resolution/dissolution of the recombination intermediates by Sgs1.Top3. PMID: 19129197
