Precision
Intra-assay Precision (Precision within an assay): CV%<8%
Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision (Precision between assays): CV%<10%
Three samples of known concentration were tested in twenty assays to assess.

Linearity
To assess the linearity of the assay, samples were spiked with high concentrations of fish LEP in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

Have you ever tested tissue sample?

A:
Thanks for your inquiry! We only tested the mentioned sample stated in the manual. We suggest you follow the manual. If you want to test tissue samplet,we suggest do pre-testing first. Here is the processing way on tissue samples.Pls check for your reference:
Tissue Homogenates 100mg tissue was rinsed with 1X PBS, homogenized in 1 ml of 1X PBS and stored overnight at -20°C. After two freeze-thaw cycles were performed to break the cell membranes, the homogenates were centrifuged for 5 minutes at 5000 x g, 2 - 8°C. The supernate was removed and assayed immediately. Alternatively, aliquot and store samples at -20°C or -80°C. Centrifuge the sample again after thawing before the assay. Avoid repeated freeze-thaw cycles.
Q:

Is this kit also compatible to test Leptin levels in Zebrafish?

A:
Thanks for your inquiry! We suggest you do a pretest if you want to test zebrafish.
Q:

Per customer request, can you please provide the following information about this kit:
a. Capture antibody or capture antigen – immunogen type, sequence (including species source and accession no. if available), expression host, clonality and host species.
b. Detection antibody (if applicable) - immunogen type, sequence (including species source and accession no. if available), expression host, clonality and host species.
c. Standard – if recombinant protein, please specify sequence (including species source and accession no. if available) and expression host.
d. Please send the manual.
e. Please provide your reference data for fish plasma samples.

A:
Thanks for your inquiry.

This is a competitive kit. There is only one antibody in the kit.
The plate is coated with mouse monoclonal antibody. ( Immunogen is CHO-derived recombinant protein.)
Standard is CHO-derived recombinant protein.
Pls find attached manual.
We tested salmon serum and plasma and some common freshwater fish before and the test value is about
125pg/ml under 1:100 dilution. The final concentration is about 12.5ng/ml.
Pls let me know if you have any further questions. Thank you.
