Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The expression region of this recombinant Human ADAMTS14 covers amino acids 253-555. The calculated molecular weight for this ADAMTS14 protein is 40.2 kDa. This protein is generated in a e.coli-based system. The ADAMTS14 coding gene included the N-terminal 6xHis tag, which simplifies the detection and purification processes of the recombinant ADAMTS14 protein in following stages of expression and purification.

The human A disintegrin and metalloproteinase with thrombospondin motifs 14 (ADAMTS14) is a member of the ADAMTS family of proteases, playing a crucial role in extracellular matrix (ECM) modulation. ADAMTS14 is involved in the cleavage of aggrecan, a major proteoglycan in the ECM of cartilage, contributing to tissue remodeling. This protease is implicated in various physiological and pathological processes, including development, inflammation, and arthritis. By cleaving ECM components, ADAMTS14 influences cell-matrix interactions and tissue homeostasis. Moreover, dysregulation of ADAMTS14 has been linked to certain diseases, highlighting its potential as a therapeutic target.

Target Background

Function
Has aminoprocollagen type I processing activity in the absence of ADAMTS2. Seems to be synthesized as a latent enzyme that requires activation to display aminoprocollagen peptidase activity. Cleaves lysyl oxidase LOX at a site downstream of its propeptide cleavage site to produce a short LOX form.
Gene References into Functions
  1. These results suggest an involvement of ADAMTS14 single-nucleotide polymorphisms rs12774070 and rs61573157 in the liver tumorigenesis and implicate the ADAMTS14 gene polymorphism as a predict factor during the progression of hepatocellular carcinoma. PMID: 28231306
  2. ADAMTS14 SNPs and environmental mutagens are a risk factor of oral tumorigenesis. PMID: 27463966
  3. Suggest the existence of an epigenetic field defect for cancerization disrupting the methylation patterns of several loci, including MGMT or ADAMTS14, that may lead to predictive biomarkers for colorectal cancer in African Americans. PMID: 25638164
  4. Data indicate that ADAMTS2, 3 and 14 cleave the amino-propeptide of fibrillar collagens. [review] PMID: 25863161
  5. No significant associations were observed in males. In conclusion, the nsSNP rs4747096 in ADAMTS14 was associated with knee OA in female Thai patients; therefore, the role of ADAMTS14 in OA seems to be gender-dependent. PMID: 24301791
