Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.

Notes

As to the activity, we haven't detect it, so we can't gurantee. The protein we produce is the full length protein, and it's purify under the mild condition. The protein has post-translational modifications and the eukaryotic expression system is more likely to be active. MP is closer to the natural state.
For the purity, our QC standard is 85% through SDS-PAGE.

Regarding the purity of 85%, I would like to know, which kind of impurities the remaining 15% are? Are these residues of the solvent or buffer or salts or other proteins?
Would a further purification by ion exchange and size exclusion chromatography be possible?

Thanks for your inquiry. The other 15% are the other proteins.
We could try the further purification. The standard can be 90%. During the further purification, it's possible the further purification failed, or the big lose of the protein. We can't guarantee. If we success, the cost will be $83, and the lead time will be 3-5 working days longer.
If we failed, we will also provide the protein with 85% purity.
For the activity, we haven't test it.

Function
Catalyzes the second step of the reductive pyrimidine degradation, the reversible hydrolytic ring opening of dihydropyrimidines. Can catalyze the ring opening of 5,6-dihydrouracil to N-carbamyl-alanine and of 5,6-dihydrothymine to N-carbamyl-amino isobutyrate.

  1. 4 newly identified DHP deficient patients presented with strongly elevated levels of 5,6-dihydrouracil and 5,6-dihydrothymine in urine and a highly variable clinical presentation, ranging from asymptomatic to infantile spasm and reduced white matter and brain atrophy. Analysis of the DHP gene (DPYS) showed the presence of 8 variants including 4 novel/rare missense variants and one novel deletion. PMID: 29054612
  2. Two unrelated pediatric DPYS deficiency cases are being described as compound heterozygotes for a novel intronic mutation c.1443+5G>A in intron 8 and a previously described missense mutation c.1001A>G (p.Q334R) in exon 6. PMID: 26771602
