PGA5 is one of the pepsinogen genes found in humans, along with PGA3 and PGA4, located on human chromosome 11 [1]. Its encoding protein pepsinogen A is an aspartic acid protease that plays a crucial role in the digestive system. It is involved in the stomach's initial stages of protein digestion by cleaving peptide bonds. Research has shown that PGA5 is expressed in Barrett's esophageal cells and its co-expression with proton pump subunits can induce cancer-associated changes [2]. Additionally, studies have highlighted the significance of PGA5 in various cancers, with differential expression patterns observed across different cancer types [3].
The expression of PGA5 has been manipulated in research settings to understand its effects. Lentiviral transduction was used to introduce PGA5 into human Barrett's esophageal cells, leading to changes associated with cancer development [2]. This manipulation technique has also been employed in creating cell lines expressing proton pump for studying conditions like laryngopharyngeal reflux and laryngeal cancer [4].
