Target Background

Function
DNA repair enzyme that cleaves apurinic/apyrimidinic (AP) sites and removes 3'-blocking groups present at single strand breaks of damaged DNA. APN1 accounts for > 97% of both apurinic/apyrimidinic (AP) endonuclease and DNA 3'-repair diesterase activities.
Gene References into Functions
  1. His-83 of Apn1 properly coordinates the active site Zn2+ ion playing a crucial role in catalytic incision stage during DNA repair. PMID: 25766873
  2. steady state levels of Apn1 V156E were substantially decreased compared to wild V156 residue plays a critical role in Apn1 stability, but is not involved in catalytic activity. PMID: 22818187
  3. Novel Apn1 substrates alpha-2'-deoxyadenosine and alpha-thymidine play an important role in counteracting oxidative DNA base damage. PMID: 15568813
  4. Apn1 is involved in an S. cerevisiae 8-oxoguanine-DNA glycosylase (Ogg1)-independent repair pathway for 8-oxo-7,8-dihydrodeoxyguanosine residues. PMID: 16024777
  5. Direct physical evidence that Apn1 not only repair cellular base damage but also prevent break accumulation that can result from abasic sites being channeled into other base excision repair pathway(s). PMID: 18267974
