Target Background

Function
Functions as ATP-binding component of the Arp2/3 complex which is involved in regulation of actin polymerization and together with an activating nucleation-promoting factor (NPF) mediates the formation of branched actin networks. Seems to contact the pointed end of the daughter actin filament.
Gene References into Functions
  1. ARP2 role in the actin filament nucleation. PMID: 27417392
  2. WASP displaces the autoinhibitory Arp3 C-terminal tail from a hydrophobic groove at Arp3's barbed end to destabilize the inactive state, providing a mechanism by which WASP stimulates the short-pitch conformation and activates the Arp2/3 complex. PMID: 27325766
  3. Studies indicate that the WASP homologue Las17 is able to nucleate actin independently of Arp2/3 through the function of G-actin-binding activities in its polyproline region. PMID: 27911716
  4. data does not support the simple hypothesis that the primary role for Arp2/3 regulators is to recruit and activate Arp2/3; rather data suggests that these regulators may be playing more subtle roles in establishing functional networks in vivo PMID: 23200048
  5. study showed that subunit ARPC1/Arc40 of the yeast actin-related protein Arp2/3 complex is indeed a stable component of the complex and contains an important contact site for the VCA domain of Wiskott-Aldrich syndrome protein family activators PMID: 15485833
  6. WASp stabilizes p35-dependent closure of the complex, holding Arp2 and Arp3 closer together to nucleate an actin filament. PMID: 15592479
  7. Binding of Arp2 with Arp3 nucleotide was found to be important for actin nucleating activity. PMID: 15657399
  8. investigated the ability of the yeast Arp2/3 complex (yArp2/3) to facilitate yeast actin polymerization in the presence and absence of the Arp2/3-activating factor Las17p WA PMID: 15857833
  9. Arp2 ATP-hydrolysis mutants exhibit defects in endocytic internalization and actin-network disassembly. PMID: 16862144
  10. These findings reveal that Arp2p also mediates what appears to be an auto-regulatory mechanism possibly adapted for efficient coordination of actin assembly and disassembly during endocytosis. PMID: 17978096
  11. An actin-related protein, Arp2, briefly colocalizes with Atg9 and directly regulates the dynamics of Atg9 movement. PMID: 18287533
  12. analysis of the key surfaces on p35/ARPC2 required for Arp2/3 complex-mediated actin assembly and cellular function PMID: 18381280
