Target Background

Function
Autophagy-specific protein that functions with ATG13, ATG29, and CIS1/ATG31 in response to autophagy-inducing signals as a scaffold to recruit other ATG proteins to organize pre-autophagosomal structure (PAS) formation. Modulates the timing and magnitude of the autophagy response, such as the size of the sequestering vesicles, through interacting with and regulating ATG1 kinase activity. Plays particularly a role in pexophagy and nucleophagy. With ATG13, is required for ATG1 activation by autophosphorylation of 'Thr-226'. Recruits ATG9 to the pre-autophagosomal structure. Plays also a role in regulation of filamentous growth.
Gene References into Functions
  1. Atg17 adopts multiple conformations in the absence of Atg31 and Atg29 suggests that the two smaller components play key roles in defining and maintaining the distinct curvature of the ternary complex PMID: 23939028
  2. single-particle electron microscopy analysis of the Atg17-Atg31-Atg29 complex reveals an elongated structure with Atg29 located at the opposing ends. PMID: 23858448
  3. The PAS scaffold proteins (Atg13 and Atg17) and phosphatidylinositol 3-kinase complex I were localized to a position at the junction between the IM and the vacuolar membrane, termed the vacuole-IM contact site (VICS). PMID: 23549786
  4. in autophagy-inducing condition, Atg9 is recruited to the pre-autophagosomal structure in a manner dependent on Atg17 PMID: 19371383
  5. Atg17-Atg13 complex formation plays an important role in normal autophagosome formation via binding to and activating the Atg1 kinase. PMID: 15743910
  6. Atg17 regulates the magnitude of the autophagic response PMID: 15901835
  7. Atg17-Atg29-Atg31 ternary complex acts as a functional unit for autophagosome formation. PMID: 19755117
